Abstract: Shp2, a protein tyrosine phosphatase possessing SH2 domains, is utilized in the intracellular signaling of various growth factors. Shp2 is highly expressed in the CNS. Brain‐derived neurotrophic factor (BDNF), a member of the neurotrophin family, which also shows high levels of expression in the CNS, exerts neurotrophic and neuromodulatory effects in CNS neurons. We examined how BDNF utilizes Shp2 in its signaling pathway in cultured cerebral cortical neurons. We found that BDNF… Expand

It is observed that Shp2 is involved in BDNF signaling in cultured cerebral cortical neurons, and this work indicates that this molecule is a positive regulator of MAPK activation and proliferation in various types of mitogenic cells. Expand
BIT/SHPS‐1 Enhances Brain‐Derived Neurotrophic Factor‐Promoted Neuronal Survival in Cultured Cerebral Cortical Neurons
TLDR
It is found that BDNF‐promoted survival of cultured cerebral cortical neurons was enhanced by expression of the wild type and also 4F mutant, indicating that this enhancement by BIT/SHPS‐1 does not depend on its tyrosine phosphorylation. Expand
Shp2 is involved in neuronal differentiation of hippocampal precursor cells
TLDR
Results suggest that Shp2 plays a positive role in mediating PDGF- and BDNF-activated signaling which promotes the formation of dendritic spines. Expand
Regulation of β‐amyloid precursor protein expression by brain‐derived neurotrophic factor involves activation of both the Ras and phosphatidylinositide 3‐kinase signalling pathways
TLDR
It is suggested that simultaneous activation of at least two signalling pathways, Ras/MAPK and PI3K/Akt, is necessary to mediate a full activation of the APP promoter by BDNF. Expand
Distinct usages of phospholipase Cγ and Shc in intracellular signaling stimulated by neurotrophins
TLDR
Different usages of phospholipase Cgamma (PLCgamma) and Shc in intracellular signaling stimulated by neurotrophins are found, indicating that neurotrophin stimulate distinct kinds of interactions between Trk and PLCGamma and between TrK and ShC, which may lead to the distinct biological functions of neurotrophs. Shp‐2 positively regulates brain‐derived neurotrophic factor‐promoted survival of cultured ventral mesencephalic dopaminergic neurons through a brain immunoglobulin‐like molecule with tyrosine‐based activation motifs/Shp substrate‐1
TLDR
Results indicated that Shp‐2 positively regulates the survival‐promoting effect of BDNF on cultured ventral mesencephalic dopaminergic neurons. Expand
Differences in survival‐promoting effects and intracellular signaling properties of BDNF and IGF‐1 in cultured cerebral cortical neurons
TLDR
There may be, as yet unidentified, intracellular signaling pathways other than the PtdIns3‐K‐Akt, MAPK and CREB signaling, to regulate survival promotion, and these unidentified signaling pathways may be responsible for the distinct strengths of the survival‐promoting effects of BDNF and IGF‐1. Expand
Inhibition of EGFR-mediated phosphoinositide-3-OH kinase (PI3-K) signaling and glioblastoma phenotype by Signal-Regulatory Proteins (SIRPs)
TLDR
Results suggest that negative regulation of PI3-K pathway activation by the SIRP family of transmembrane receptors may diminish EGFR-mediated motility and survival phenotypes that contribute to transformation of glioblastoma cells. Expand
Hepatocyte Growth Factor-induced Differential Activation of Phospholipase Cγ1 and Phosphatidylinositol 3-Kinase Is Regulated by Tyrosine Phosphatase SHP-1 in Astrocytes*
TLDR
It is shown that HGF promoted chemokinesis of rat primary astrocytes through the activation of phosphatidylinositol 3 (PI3)-kinase without any influence on mitogenesis of the cells and that PLCγ1 is selectively prevented from being a signal mediator by constitutive association of SHP-1. Expand

N-Shc: a neural-specific adapter molecule that mediates signaling from neurotrophin/Trk to Ras/MAPK pathway.
TLDR
The isolation of a neural-specific member of the Shc family, named N-Shc (neuronal Shc), contains two potential phosphotyrosine-binding domains, PTB and SH2, and is expressed exclusively in the brain; whereas Shc is present in all other non-NEuronal tissues. Expand

Transient Association of the Phosphotyrosine Phosphatase SHP‐2 with TrkA Is Induced by Nerve Growth Factor
TLDR
This is the first demonstration that the association of SHP‐2 with TrkA is induced by NGF and that this association leads to SHp‐2 activation and tyrosine phosphorylation, and it is concluded that SHP-2 plays a significant role in early biochemical events in Trk a‐mediated signal transduction.
