The enzymic self-polymerization of prophobilinogen gives rise to the cyclic tetrapyrroles uroporphyrinogen III and uroporphyrinogen I. The former is the precursor of all the natural porphyrins and chlorins. The formation of uroporphyrinogen III is catalysed by a dual enzymic system, porphobilinogen deaminase and uroporphyrinogen III cosynthase. Deaminase polymerizes four porphobilinogen units on the enzymic surface, without liberation of free intermediates into the reaction medium, and forms… Expand

Porphobilinogen deaminase and uroporphyrinogen III synthase: Structure, molecular biology, and mechanism
The human deaminase structure has been modeled from the E. coli structure and has led to a molecular explanation for the disease acute intermittent porphyria and molecular modeling has been employed to simulate the spiro-mechanism of uroporphyrinogen III synthase. Expand
Biosynthesis of uroporphyrinogens. Interaction among 2-(aminomethyl)bilanes and the enzymatic system.
An outline of the most likely pathway of uroporphyrinogen III biosynthesis from porphobilinogen is given and results obtained with 2-aminomethyldipyrrylmethanes are obtained. Expand
Spectroscopic evidence for a porphobilinogen deaminase-tetrapyrrole complex that is an intermediate in the biosynthesis of uroporphyrinogen III.
While the HMB inhibited uro'gen III formation from 2-(hydroxymethyl)bilane (HMB) and from the deaminase-tetrapyrrole intermediate was compared by using deamin enzyme-cosynthase and cosynth enzyme from several sources, it was found that while the H MB inhibited uroporphyrinogen I formation at higher concentrations and longer incubation times, uro’gen III form did not decrease with time. Porphyrin biosynthesis in Euglena gracilis—II. Pyrrylmethane intermediates in the enzymic cyclotetramerization of porphobilinogen
Abstract 1. 1. Under normal incubation conditions pellet and deaminase Euglena gracilis preparations, are able to form two different polypyrrole intermediates called P and D, respectively, which didExpand
Structural basis of pyrrole polymerization in human porphobilinogen deaminase.
Human porphobilinogen deaminase (PBGD), the third enzyme in the heme pathway, catalyzes four times a single reaction to convert porphobilinogen into hydroxymethylbilane. Remarkably, PBGD employs aExpand
Chapter 1 The biosynthesis of 5-aminolaevulinic acid and its transformation into uroporphyrinogen III
This chapter discusses the enzymic synthesis of 5-aminolaevulinic acid and the subsequent three enzyme steps to porphobilinogen, preuroporphyrinogen and uroporphyrinogens III, common to all living systems which synthesize tetrapyrroles. Expand
The biosynthesis of haem in congenital (erythropoietic) porphyria.
It is confirmed that the primary control of haem biosynthesis in congenital porphyria is at the level of δ-aminolaevulinic acid synthase with a secondary control point at thelevel of porphobilinogen deaminase. Expand

It is assumed that porphobilinogen deaminase is essential for the synthesis of corrinoids, and both the activity of ribonucleotide reductase and the amount of vitamin B12 were significantly lower in the first group. Expand
Synthesis of porphobilinogen‐9‐14C
Ethyl 3-dimethylaminomethyl-5-benzyloxy-6-azaindole-2-carboxy-late was condensed with the sodium salt of diethyl malonate-2-14C in decaline, and ethyl 5-benzyloxy-2-ethoxy carbonyExpand
Evidence is presented that each of the porphyrias represents a different inborn error of metabolism in haem biosynthesis, and it is suggested that insituations where the activity of this enzyme is derepressed, prophobilinogen deaminase represents a secondary control step. Expand

Polypyrroles formed from porphobilinogen and amines by uroporphyrinogen synthetase of Rhodopseudomonas spheroides.
The behaviour, properties and composition of the radioactive hydroxylamine derivative were consistent with a tetrapyrrolic structure, probably a pyrrylmethane, that was not cyclized, rather than with di-, tri- or penta-pyrro Lic structures. Expand
Studies on porphobilinogen deaminase and uroporphyrinogen 3 cosynthase from human erythrocytes.
Deaminase and deaminase-cosynthase showed Michaelian kinetics and the whole complex formed uroporphyrinogen III, which was found to be inactivated by photooxidation and chemical oxidations, by sulphydryl reagents and by divalent metals. Expand
Studies on the porphobilinogen deaminase-uroporphyrinogen cosynthetase system of cultured soya-bean cells.
Porphobilinogenase activity was affected by the presence of air; uroporphyrinogens were only formed under anaerobic conditions, although substrate consumption was the same in the absence of oxygen as in its presence and a sharp optimum at pH 7.2 was obtained. Expand
Biosynthesis of porphyrins and related macrocycles. I. Synthesis of 14C-labelled pyrromethanes.
R Routes are developed as follows for rational synthesis of the labelled (aminomethyl)pyrromethanes (6) and (7), which are of importance for incorporation studies on haem, protoporphyrin-IX, and vitamin B12. Expand
The enzymatic inactivation of porphobilinogen deaminase
Abstract 1. 1. Porphobilinogen deaminase was inhibited by pyrrolooxygenase, a mixed-function oxidase which inhibits tryptophan-containing enzymes in the presence of sodium dithionite. 2. 2. BothExpand
Purification and properties of porphobilinogen deaminase from wheat germ.
Porphobilinogen deaminase from wheat germ was purified about 1000-fold and a dithionite-activated protein factor was found to consume PBG in a special way and to interact with the PBG-deaminase by inhibiting its activity. Expand
