Sphingosine kinase (SphK) 1 and 2 phosphorylate sphingosine to generate sphingosine-1-phosphate (S1P), a pluripotent lipophilic mediator implicated in a variety of cellular events. Here we show that the activity of β-site APP cleaving enzyme-1 (BACE1), the rate-limiting enzyme for amyloid-β peptide (Aβ) production, is modulated by S1P in mouse neurons. Treatment by SphK inhibitor, RNA interference knockdown of SphK, or overexpression of S1P degrading enzymes decreased BACE1 activity, which… Expand
Reduced sphingosine kinase-1 and enhanced sphingosine 1-phosphate lyase expression demonstrate deregulated sphingosine 1-phosphate signaling in Alzheimer’s disease
TLDR
The importance of S1P in AD is highlighted suggesting the existence of a global deregulation of S 1P signaling in this disease from its synthesis by SphK1 and degradation by SPL to its signaling by the S1p1 receptor. Expand
Sphingosine Kinases/Sphingosine-1-Phosphate and Death Signalling in APP-Transfected Cells
TLDR
It is shown that Aβ peptides evoke down-regulation of gene expression and activity for SphK(s) and S1P1 and that the cytoprotective effect of S1p was receptor-independent. Expand
Sphingosine-1-phosphate signaling and its role in disease.
The bioactive sphingolipid metabolite sphingosine-1-phosphate (S1P) is now recognized as a critical regulator of many physiological and pathophysiological processes, including cancer,Expand
Neuronal sphingosine kinase 2 subcellular localization is altered in Alzheimer’s disease brain
TLDR
The results suggest that a shift in the subcellular localization of the S1P generating SphK2 may compromise the well established pro-survival cytosolic S 1P by favoring the production of nuclear S1p associated with adverse effects in AD pathogenesis. Expand

TLDR
FTY720 and KRP203 decreased production of Amyloid-β peptide (Aβ), a pathogenic proteins causative for Alzheimer disease (AD), in cultured neuronal cells, suggesting that S1PR modulators are novel type of regulators for Aβ metabolisms that are active in vitro and in vivo. Expand

Distribution of sphingosine kinase activity and mRNA in rodent brain
TLDR
Results indicate that SPHK2 is the predominant S1P‐synthesizing isoform in normal brain parenchyma, and its heterogeneous distribution, in particular laminar distribution in cortex, suggests a neuronal localization and a possible role in cortical and cerebellar functions, in normal as well as ischemic brain. Pharmacology and Antitumor Activity of ABC294640, a Selective Inhibitor of Sphingosine Kinase-2
TLDR
Oral administration of ABC294640 to mice bearing mammary adenocarcinoma xenografts results in dose-dependent antitumor activity associated with depletion of S1P levels in the tumors and progressive tumor cell apoptosis, and this newly developed SK2 inhibitor provides an orally available drug candidate for the treatment of cancer and other diseases.
