The present study was undertaken to investigate the autoinhibition and desensitization of 5-hydroxytryptamine (5-HT) using another agonist MK-212 on guinea pig ileum. 5-HT and MK-212 produced dose dependent contractions of guinea pig ileum. The responses to MK-212 were reduced in the presence of 5-HT and vice versa. Neither 5-HT nor MK-212 produced any change in the responses to histamine, acetylcholine or KCl. Increase in Ca++ in bathing fluid reversed the desensitization produced by MK-212 or… Expand

Dual agonist-antagonist effects of 5-hydroxytryptamine (5-HT) in the guinea pig ileum: evidence for a selective receptor desensitization effect.
TLDR
Results suggest that fade and the auto-blockade are part of a common effect and are discussed in relation to a model of drug-induced selective receptor desensitization. Expand
A PHARMACOLOGICAL ANALYSIS OF THE MODE OF ACTION OF SEROTONIN (5‐HYDROXYTRYPTAMINE) UPON THE GUINEA‐PIG ILEUM
TLDR
The provisional conclusion is drawn that serotonin acts upon the post-ganglionic cholinergic fibres of the intramural nervous system of the guinea-pig ileum, and is responsible for the stimulation of the gut and increased sensitivity to other drugs. Expand
TWO KINDS OF TRYPTAMINE RECEPTOR
TLDR
There are two kinds of tryptamine receptor in the guinea‐pig ileum, namely the M receptors which can be blocked with morphine and the D receptors which have no additional effect after dibenzyline. Expand
Central serotonin-like activity of 6-chloro-2-[1-piperazinyl]-pyrazine (CPP; MK-212).
TLDR
Findings indicate that CPP has a serotoninmimetic action in the central nervous system, similar to that produced by pargyline plus tryptophan or other treatments effecting increased activation of central serotonin receptors. Expand

TLDR
The central focus in this review will be on the sequence of cellular reactions that appear to be essen tial for linking the interaction of the drug and receptor to the activation of the contractile apparatus. Expand
A 5‐HYDROXYTRYPTAMINE‐LIKE MODE OF ANORECTIC ACTION FOR 6‐CHLORO‐2‐[1‐PIPERAZINYL]‐PYRAZINE (MK‐212)
TLDR
The results indicate that the anorectic action of MK‐212 involves a 5‐HT‐like component which is more evident at the higher dose level of the compound, and may depend, at least partly, upon the integrity of 5‐ HT‐containing neurones in the central nervous system. Expand
