It is increasingly clear that asthma is a complex disease made up of number of disease variants with different underlying pathophysiologies. Limited knowledge of the mechanisms of these disease subgroups is possibly the greatest obstacle in understanding the causes of asthma and improving treatment and can explain the failure to identify consistent genetic and environmental correlations to asthma. Here we describe a hypothesis whereby the asthma syndrome is divided into distinct disease… Expand

  • Current opinion in pulmonary medicine
  • 2016
TLDR
The review describes the current understanding of how asthma phenotypes and endotypes can be employed to gain insight into asthma pathobiology and personalized therapy. Expand
Childhood and Adult Asthma: Phenotype- and Endotype-Based Biomarkers
TLDR
This book chapter aims to discuss the current concepts on asthma classification and biomarker-based diagnosis and may in future constitute a paradigm shift from the traditional pharmacologic treatments and lead to better prognosis in moderate-to-severe forms of the disease. Expand
Clinical entities, phenotypes, causation, and endotypes based on selected asthma publications
TLDR
Focusing on causation can improve the understanding of disease entities, disease classification, and disease causation in nearly all diseases. Expand
Asthma Phenotypes and Endotypes: Implications for Personalised Therapy
TLDR
The role of functional imaging, physiology, blood-, sputum- and breath-based biomarkers and clinical manifestations that could be used to produce a personalised asthma profile are considered, with implications on prognosis, pathophysiology and most importantly specific therapeutic responses. Expand
Untangling asthma phenotypes and endotypes
TLDR
Whether newly defined asthma endotypes predict the individual course of asthma has to be validated in longitudinal studies and the identification of corresponding molecular biomarkers for individual pathogenic mechanism underlying phenotypes or subgroups within a phenotype is important. Expand
Phenotypes and endotypes of adult asthma: Moving toward precision medicine.
TLDR
Advances in the understanding of the diverse immunologic perturbations that drive airway inflammation are consistent with clinical traits associated with these phenotypes and their response to biologic therapies, and these disease subtypes represent asthma endotypes. Expand
Asthma phenotypes and endotypes: an evolving paradigm for classification.
TLDR
A growing understanding of these mechanistically distinct groups, along with the identification of relevant cellular or molecular biomarkers, is already showing promise as a way of predicting clinical response to various asthma therapies. Expand
Phenotypes and endotypes of uncontrolled severe asthma: new treatments.
TLDR
A review of the most relevant aspects of the clinical and inflammatory phenotypes of severe asthma, including severe childhood asthma and the various endotypes, and the main therapeutic options available for patients with uncontrolled severe asthma. Expand
Asthma phenotypes and endotypes
TLDR
The present understanding of different asthma phenotypes and endotypes, and the prospects of personalized medicine for asthma are discussed, as well as the effectiveness of tailoring asthma treatment on the basis of an individual's biology. Expand
The concepts of asthma endotypes and phenotypes to guide current and novel treatment strategies
TLDR
The definition of asthma phenotypes and possible endotypes, advances in allergy-immunology field and contemporary personalized therapy options for asthma are reviewed, indicating a new era termed as ‘precision medicine’. Expand

Endotyping asthma: new insights into key pathogenic mechanisms in a complex, heterogeneous disease
TLDR
This approach is already suggesting entirely novel pathways to disease-eg, alternative macrophage specification, steroid refractory innate immunity, the interleukin-17-regulatory T-cell axis, epidermal growth factor receptor co-amplification, and Th2-mimicking but non-T-cell,interleukins 18 and 33 dependent processes that can offer unexpected therapeutic opportunities for specific patient endotypes.
