Drug-induced liver injury (DILI) is caused by unpredictable adverse drug reaction due mainly to the accumulation of hepatotoxic compounds in the liver resulting in significant damage. Drug-metabolizing enzymes have been prime targets for molecular studies relevant to DILI. The gene UGT1A9 mainly expresses in the liver and has an important role in drug metabolism. The Han Chinese has a very long and complex demographic history, and the population stratification arising from the interplay of… Expand
Association between genetic polymorphisms of SLCO1B1 and susceptibility to methimazole‐induced liver injury
The results suggested that genetic polymorphisms of SLCO1B1 were associated with susceptibility to MMI‐DILI, and may be important predisposing factors for M MI‐induced hepatotoxicity. Expand
Genome‐Wide Analysis of DNA Methylation and Antituberculosis Drug‐Induced Liver Injury in the Han Chinese Population
A CRISPR/Cas9‐mediated methylation modifiable cell model was applied and it was demonstrated that four CpGs in or near the gene region of AK2, SLC8A2, and PSTPIP2 affected the cellular response to rifampicin treatment. Expand
Epidemiology and Genetic Risk Factors of Drug Hepatotoxicity.
Genetic analysis of DILI is currently limited, but multiple polymorphisms of human leukocyte antigen genes and genes involved in drug metabolism and transport have been identified as risk factors. Expand
Proteome and transcriptome analysis revealed florfenicol via affected drug metabolism and lipid metabolism induce liver injury of broilers
The results showed that FFC can affect the drug metabolism and lipid synthesis in the liver of broiler, thus impairing the normal function of liver and the growth and development of broilers. Expand
Drug-Induced Liver Injury: Highlights from a Review of the 2015 Literature
New DILI biomarkers, specifically microRNA-122 and keratin-18, appear to have the necessary predictive value to determine the prognosis and outcome of patients with paracetamol-induced acute liver failure (ALF), and may be of great benefit in deciding who requires N-acetylcysteine (NAC), and for what duration. Regorafenib induced severe toxic hepatitis: characterization and discussion
Regorafenib is the first small‐molecule multikinase inhibitor which showed survival benefits in pretreated metastatic colorectal cancer (mCRC) patients. Besides classical adverse events of this drugExpand

Identification of common polymorphisms in the promoter of the UGT1A9 gene: evidence that UGT1A9 protein and activity levels are strongly genetically controlled in the liver.
This is the first study to demonstrate that naturally occurring sequence variations in the UGT1A9 promoter are informative in predicting the levels of protein and glucuronidating activity, providing a potential mechanism for interindividual variation in UGT 1A9-mediated metabolism. Expand
Genetic polymorphism analysis of cytochrome P4502E1 (CYP2E1) in Chinese Han populations from four different geographic areas of Mainland China.
Twenty-two different CYP2E1 polymorphisms, including six novel variants in promoter regions and a novel nonsense mutation, were identified and the frequencies of some polymorphisms and genotypes demonstrated significant differences among the four populations. Expand
Genetic polymorphism analysis of CYP2C19 in Chinese Han populations from different geographic areas of mainland China.
This is the first study that systematically screened the polymorphisms of the whole CYP2C19 gene in a large Chinese Han population and suggests that a few low frequent variants show significant differences among the four populations, while the prevalent polymorphisms show no differences. Expand
Systematic screening for polymorphisms within the UGT1A6 gene in three Chinese populations and function prediction through structural modeling.
The study suggests that genetic polymorphisms in UGT1A6 may contribute to interindividual and intra-ethnic differences and should prove helpful in the development of pharmacogenomics in China. Expand
Genetic susceptibility to diclofenac-induced hepatotoxicity: contribution of UGT2B7, CYP2C8, and ABCC2 genotypes.
Allelic variants of UGT2B7, CYP2C8, and ABCC2, which may predispose to the formation and accumulation of reactive diclofenac metabolites are associated withdicl ofenac hepatotoxicity. Expand
A systematic genetic polymorphism analysis of the CYP2C9 gene in four different geographical Han populations in mainland China.
This study used direct sequencing to systematically screen genetic polymorphisms of the CYP2C9 gene including the 5' -flanking region (2kb), all exons and their adjoining intron regions and the 3' UTR in 400 unrelated healthy Chinese Han volunteers. Expand
A Systematically Combined Genotype and Functional Combination Analysis of CYP2E1, CYP2D6, CYP2C9, CYP2C19 in Different Geographic Areas of Mainland China – A Basis for Personalized Therapy
This study is the first to systematically analyze the combined genotypes and functional combinations of 400 samples of major CYP genes in four geographical areas of mainland China and could make a significant contribution in the field of personalized medicine and therapy. Expand
Cytochrome P450 variations in different ethnic populations
Racial/ethnic differences in metabolic phenotype can be explained by differences in SNP distribution, but overlap in substrate specificity, linkage disequilibrium and previously unidentified SNPs have made phenotypic characterization difficult for CYP3A4/5 and 2C8/9. Expand
Genotype and allele frequencies of polymorphic UGT1A9 in the Polish population
It is demonstrated that the frequency of the variant UGT1A9*3 was 0.016, which suggests the need for detailed analysis of its effect on important drugs metabolism level in Polish population, and finds frequencies of these genetic variations in 308 healthy individuals representing Polish population.
