MY-1, a fraction extracted from BCG and composed of 70.0% DNA and 28.0% RNA, was examined for its antitumor activity against 9 different syngeneic mouse tumors. Tumor regression was induced in almost all of the mice bearing any of five kinds of solid tumors by repeated intralesional injections of 100 micrograms MY-1. When cells of some tumors were inoculated intradermally together with MY-1, tumor growth was suppressed, lung metastases were inhibited, and the survival times of mice bearing 1 of…

Results indicate that intratumoral MY-1 injections induce a MethA-specific, L3T4-positive cell-mediated, delayed-type hypersensitivity, which is necessary for the tumor regression.
Activation of NK cell (human and mouse) by immunostimulatory DNA sequence

Antitumor activity of MY-1 was also abolished if the animals were pretreated with asialo GM1 antiserum or carrageenan suggesting that the activity can be ascribed mainly to activated NK cells [16].

It was demonstrated that repeated intralesional injections of the complex resulted in delay of tumor growth and complete cure of animals from L10 tumor inoculated, and similar treatment of the animals with the same amount of MY-1 or poly LL/CMC alone had little therapeutic effect on the tumor growth.

The response of higher animals against immunostimulatory DNA must be the most primitive but important mechanism for self-nonself discrimination against foreign DNA.
Antimetastatic effect of CpG DNA mediated by type I IFN.
- Biology, MedicineCancer research

Data indicate that CpG-ODNs activate NK/NKT cells for antimetastatic activity indirectly via IFN-alpha/beta receptor activation, and the exploitation of the stimulatory activity of Cpg-ODN for the innate immune system might be a useful strategy for antimETastatic therapy.

- Biology, MedicineSpringer Seminars in Immunopathology

DNA purified from BCG inhibited the growth of various animal tumors, augmented cell activity and induced IFN from mouse spleen cells, and the response of higher animals against immunostimulatory DNA must be the most primitive but important mechanism for self-nonself discrimination against foreign DNA.
