The antinociceptive properties of the hydro-methanolic extract (HME) and two flavonoids isolated fromDanae racemosa have been investigated in several nociceptive rat models. The HME fromD. racemosa (100-400 mgkg-1, i.p.) produced significant dose-related inhibition of acetic acid-induced abdominal constriction. In the same dose range, the HME produced dose-related inhibition in both phases of a formalin-test. Treatment of animals with naloxone (5 mgkg-1, i.p.) completely reversed the…

It is concluded that HE exhibits a potent antinociceptive profile, either when given intraperitoneally or orally, and appears to be unrelated to inhibition of synthesis of arachidonic acid via cyclo‐oxygenase or to activation of opioid receptors.

Its antinociceptive effect was insensitive to naloxone, suggesting the lack of involvement of endogenous opioid, was not modulated by adrenal glands and does not involve interaction with the L-arginine-nitric oxide pathway, activation of alpha-1 adrenoceptors or tau-aminobutyric acidB receptors, but requires, at least in part, the serotoninergic pathway.
