Antibodies are important tools for a broad range of applications due to their high specificity and ability to recognize virtually any target molecule. However, in order to be practically useful, antibodies must be highly stable and bind their target antigens with high affinity. We present a combinatorial approach to generate high-affinity, highly stable antibodies through the design of stable frameworks, specificity grafting and maturation via somatic hypermutation in vitro. By collectively…

Figures and Tables from this paper

A general approach to antibody thermostabilization
TLDR
Antibody engineering to enhance thermostability may be broadly useful for improving the biophysical characteristics of antibodies across a number of applications.
Enhanced stabilization of a stable single domain antibody for SEB toxin by random mutagenesis and stringent selection.
TLDR
This work utilizes random mutagenesis and stringent selection in an effort to obtain variants of A3 with even higher melting points, and demonstrates that by the addition of relatively subtle changes it is possible to further improve the melting temperature of single domain antibodies that are already remarkably stable.
Toward Drug-Like Multispecific Antibodies by Design
TLDR
Advances in the design and engineering of multispecific antibodies with drug-like properties, including favorable stability, solubility, viscosity, specificity and pharmacokinetic properties are reviewed.
Comprehensive optimization of a single-chain variable domain antibody fragment as a targeting ligand for a cytotoxic nanoparticle
TLDR
It is demonstrated that tyrosine residues in the CDRs make diverse contributions to the binding affinity and biophysical properties, and that replacement of non-essential tyrosines can improve the stability and bioactivity of the scFv.
Improving antibody thermostability based on statistical analysis of sequence and structural consensus data
TLDR
An inhouse application for predicting human MAb thermostability to guide protein engineers to design more stable molecules is developed and has shown success in designing MAb engineering panels in multiple biologics programs.
Efficient affinity maturation of antibody variable domains requires co-selection of compensatory mutations to maintain thermodynamic stability
TLDR
It is demonstrated that the accumulation of both affinity and stability mutations is necessary to maintain thermodynamic stability during extensive mutagenesis and affinity maturation in vitro, which is similar to findings for natural antibodies that are subjected to somatic hypermutation in vivo.
Contributions of the Complementarity Determining Regions to the Thermal Stability of a Single-Domain Antibody
TLDR
Results from the CDR swap mutants indicate CDR interactions play a major role in the protein stability and showed that CDR2 plays a critical role in sdAb A3’s binding and stability.
Solubility, Stability, and Avidity of Recombinant Antibody Fragments Expressed in Microorganisms
TLDR
Attempts on not only the production of antibodies or antibody fragments in microorganisms but also scFv stabilization via directed evolution of variants with increased stability using display systems, and stabilization of the interface between variable regions of heavy (VH) and light (VL) chains through the introduction of a non-native covalent bond between the two chains are discussed.

Improving in vivo folding and stability of a single-chain Fv antibody fragment by loop grafting.
TLDR
The complementary determining regions (CDRs) from the fluorescein-binding antibody 4-4-20, which yields almost no soluble protein in periplasmic expression in Escherichia coli, were transplanted to the framework of the humanized antibody 4D5 and showed both a dramatic improvement in soluble expression and improved thermodynamic stability.
Stabilization of antibody fragments in adverse environments
TLDR
It has been found that a novel antibody fragment construct (STAB: stabilized antibody) containing both a flexible linker and a disulphide bond can be effectively produced and shows greatly improved stability in these diverse environments.
Transfer of engineered biophysical properties between different antibody formats and expression systems.
TLDR
The results confirm the potential of structure-based protein engineering in the context of full-length IgGs and the transferability of stability improvements discovered with smaller antibody fragments, and indicate that engineered mutations can increase functional expression levels only for periplasmic expression in prokaryotes.
A broad range of Fab stabilities within a host of therapeutic IgGs.
Selecting and screening recombinant antibody libraries
TLDR
The first antibody of this new generation, adalimumab (Humira, a human IgG1 specific for human tumor necrosis factor (TNF)), already approved for therapy and with many more in clinical trials, these recombinant antibody technologies will provide a solid basis for the discovery of antibody-based biopharmaceuticals, diagnostics and research reagents for decades to come.
Affinity enhancement of an in vivo matured therapeutic antibody using structure‐based computational design
TLDR
The results indicate that structure‐based computational design can be successfully applied to further improve the binding of high‐affinity antibodies and improve the single‐mutant success rate.
Consensus-based engineering of protein stability: from intrabodies to thermostable enzymes.
Coupling mammalian cell surface display with somatic hypermutation for the discovery and maturation of human antibodies
TLDR
This approach overcomes many of the previous limitations of mammalian cell display, enabling direct selection and maturation of antibodies as full-length, glycosylated IgGs.
