Physical or chemical disruption of the structure of liver mitochondria usually leads to preparations with considerable respiratory activity, but with little or no activity in coupling phosphorylation to respiration. Under carefully controlled conditions, however, submitochondrial preparations with considerable phosphorylating activity may be obtained (l-5).
