We report an alternative, hydroxylating pathway for the metabolism of vitamin D2 in a cytochrome P450 side chain cleavage (P450scc; CYP11A1) reconstituted system. NMR analyses identified solely 20‐hydroxyvitamin D2 and 17,20‐dihydroxyvitamin D2 derivatives. 20‐Hydroxyvitamin D2 was produced at a rate of 0.34 mol·min−1·mol−1 P450scc, and 17,20‐dihydroxyvitamin D2 was produced at a rate of 0.13 mol·min−1·mol−1. In adrenal mitochondria, vitamin D2 was metabolized to six monohydroxy products… Expand

Purified Mouse CYP27B1 Can Hydroxylate 20,23-Dihydroxyvitamin D3, Producing 1α,20,23-Trihydroxyvitamin D3, Which Has Altered Biological Activity
TLDR
It is shown that purified CYP27B1 can add a 1α-hydroxyl group to 20,23(OH)2D3 with the product showing altered biological activity, especially for the stimulation of CYP24 gene expression. Expand
Hydroxylation of CYP11A1-Derived Products of Vitamin D3 Metabolism by Human and Mouse CYP27B1
TLDR
CYP11A1-derived secosteroids are 1α-hydroxylated by both human and mouse CYP27B1 with low catalytic efficiency, and that the presence of a 17α,20-Dihydroxyvitamin D3 group completely blocks 1 α-Hydroxylation. Expand
Detection of 7-Dehydrocholesterol and Vitamin D3 Derivatives in Honey
TLDR
The presence of 7DHC and D3 and of species corresponding to 20(OH)-7DHC, 1,20-dihydroxyvitamin D3, 25(OH)D3 and 1,25( OH)2D3 in honey implies their production in bees, although the precise biochemistry and photochemistry of these processes remain to be defined. Expand
The serum vitamin D metabolome: What we know and what is still to discover
TLDR
With advances in LC/MS/MS and its increased availability, and with the help of studies with recombinant vitamin D-metabolizing enzymes, many other vitamin D metabolites have now been detected and in some cases quantitated, in human serum, some contributing to a more detailed understanding of vitamin D status in health and disease. Expand
Endogenously produced nonclassical vitamin D hydroxy-metabolites act as “biased” agonists on VDR and inverse agonists on RORα and RORγ
TLDR
It is suggested that the identification of large number of endogenously produced alternative hydroxy-metabolites of D3 that are biologically active, and of possible alternative receptors, may offer an explanation for the pleiotropic and diverse activities of vitamin D, previously assigned solely to 1,25(OH)2D3 and VDR. Expand
Photoprotective Properties of Vitamin D and Lumisterol Hydroxyderivatives
TLDR
The protection of human keratinocytes against DNA damage included the activation of the NRF2-regulated antioxidant response, p53-phosphorylation and its translocation to the nucleus, and DNA repair induction, indicating that novel derivatives of vitamin D3 and lumisterol are promising photoprotective agents. Expand
Novel non-calcemic secosteroids that are produced by human epidermal keratinocytes protect against solar radiation
TLDR
Novel secosteroids that can be produced by the action of CYP11A1 in epidermal keratinocytes have protective effects against UVB radiation. Expand
Antifibrogenic activities of CYP11A1-derived vitamin D3-hydroxyderivatives are dependent on RORγ.
TLDR
The results show that the antiproliferative and antifibrotic activities of the vitamin D hydroxy-derivatives are dependent on a functional RORγ. Expand
The significance of CYP11A1 expression in skin physiology and pathology
TLDR
CYP11A1 catalyzes the first and rate-limiting step in steroidogenesis, converting cholesterol to pregnenolone, and plays several critical roles in the skin through its initiation of local steroidogenesis and specific metabolism of vitamin D, lumisterol, and 7-dehydrocholesterol. Expand

The cytochrome P450scc system opens an alternate pathway of vitamin D3 metabolism
TLDR
It is concluded that purified P450scc in a reconstituted system or P450Scc in adrenal mitochondria can add one hydroxyl group to vitamin’D3 with subsequent hydroxyation being observed for reconst ituted enzyme but not for adrenAL mitochondria. Expand

TLDR
7-DHC can form 7-DHP through P450scc side-chain cleavage, which may serve as a substrate for further conversions into hydroxy derivatives through existing steroidogenic enzymes, and has the potential to generate a variety of molecules depending on local steroidogenic activity and access to UVB.
