Juvenile rats are more susceptible than adults to the acute toxicity of organophosphorus insecticides like chlorpyrifos (CPF). Age- and dose-dependent differences in metabolism may be responsible. Of importance are CYP450 activation and detoxification of CPF to chlorpyrifos-oxon (CPF-oxon) and trichloropyridinol (TCP), as well as B-esterase (B-est) and PON-1 (A-esterase) detoxification of CPF-oxon to TCP. In the current study, a physiologically based pharmacokinetic/pharmacodynamic (PBPK/PD…
Sensitivity analysis on a physiologically-based pharmacokinetic and pharmacodynamic model for diisopropylfluorophosphate-induced toxicity in mice and rats
TLDR
A physiologically-based pharmacokinetic and pharmacodynamic (PBPK/PD) model developed to study the effect of diisopropylfluorophosphate (DFP) on acetylcholinesterase (AChE) activity in mouse and rat identified parameters that contribute most to the variability of AChE dynamics for model optimization against data.
Chlorpyrifos PBPK/PD model for multiple routes of exposure
TLDR
A physiologically based pharmacokinetic/pharmacodynamic (PBPK/PD) model has been developed that describes the relationship between oral, dermal and inhalation doses of CPF and key events in the pathway for cholinergic effects.
A physiologically based pharmacokinetic model for quinoxaline-2-carboxylic acid in rats, extrapolation to pigs.
TLDR
A multi-compartment physiologically based pharmacokinetic (PBPK) model to describe the disposition of cyadox and its metabolite quinoxaline-2-carboxylic acid after a single oral administration in rats supported the validity of the model with simulated tissue concentrations within the range of the observations.
Carbaryl and 1-Naphthol Tissue Levels and Related Cholinesterase Inhibition in Male Brown Norway Rats from Preweaning to Senescence
TLDR
Tissue levels of the N-methyl carbamate carbaryl and its metabolite 1-naphthol were determined and related to brain and RBC cholinesterase (ChE) inhibition in the same animals, and there was more variability in the relationship between RBC ChE and plasma carbaryl levels.

A Physiologically based pharmacokinetic and pharmacodynamic (PBPK/PD) model for the organophosphate insecticide chlorpyrifos in rats and humans.
TLDR
The model quantitatively estimates target tissue dosimetry and AChE inhibition and is a strong framework for further organophosphate (OP) model development and for refining a biologically based risk assessment for exposure to CPF under a variety of scenarios.
Age-related differences in parathion and chlorpyrifos toxicity in male rats: target and nontarget esterase sensitivity and cytochrome P450-mediated metabolism.
TLDR
Results indicate that target enzyme sensitivity is not responsible for age-related toxicity differences, nor is the potential for hepatic bioactivation, whereas lower levels of hepatic aliesterase-mediated protection and P450-mediated dearylation probably contribute significantly to the greater sensitivity of juveniles to phosphorothionate toxicity.
In vitro rat hepatic and intestinal metabolism of the organophosphate pesticides chlorpyrifos and diazinon.
TLDR
Investigating the role intestinal and hepatic metabolism may play in both the activation and detoxification of CPF and DZN in Sprague-Dawley rats suggests that intestinal metabolism may impact the metabolism of CPf and DN, especially following low-dose oral exposures.
Physiologically based pharmacokinetic and pharmacodynamic model for the inhibition of acetylcholinesterase by diisopropylfluorophosphate.
TLDR
A physiologically based model for diisopropylfluorophosphate (DFP) pharmacokinetics and acetylcholinesterase (AChE) inhibition was developed in mammals and demonstrated the possibility of using a model based on mammalian physiology and biochemistry to simulate in vivo data on DFP pharmacokinetic and AChE inhibition.
Tissue-specific effects of chlorpyrifos on carboxylesterase and cholinesterase activity in adult rats: an in vitro and in vivo comparison.
TLDR
There are tissue and gender-related differences for basal ChE and CaE activity and the in vitro sensitivity of CaE or ChE to CPF-oxon is highly tissue-specific, suggesting that the number of binding sites does play a role in the detoxification potential of a tissue.
Age-related brain cholinesterase inhibition kinetics following in vitro incubation with chlorpyrifos-oxon and diazinon-oxon.
TLDR
An age- and OP-selective inhibition of rat brain ChE is demonstrated, which may be critically important in understanding the potential sensitivity of juveniles to specific OPs exposures.
