}The Na(+)-K(+)-Cl(-) cotransporters (NKCCs), which belong to the cation-Cl(-) cotransporter (CCC) family, are able to translocate NH4(+) across cell membranes. In this study, we have used the oocyte expression system to determine whether the K(+)-Cl(-) cotransporters (KCCs) can also transport NH4(+) and whether they play a role in pH regulation. Our results demonstrate that all of the CCCs examined (NKCC1, NKCC2, KCC1, KCC3, and KCC4) can promote NH4(+) translocation, presumably through binding…

Phosphoregulation of K+‐Cl− cotransporter 4 during changes in intracellular Cl− and cell volume
- BiologyJournal of cellular physiology

It is shown for the first time that one of the KCCs can be regulated through direct phosphorylation, that changes in [Cl−]i or CVOL modify the activity of signaling enzymes at carrier sites, and that the effectors directly involved do not include a Cal A‐sensitive PP in contrast to the widely held view.

There are still a number of uncertainties regarding the transport properties, structural organization and regulation of KCC3, but it has become obvious that each one of these isoforms exhibits unique patterns of distribution and fulfill distinct physiological roles.

KCC1 has been found to be the most widely distributed KCC isoform and considered to act as a housekeeping membrane protein, which appears to play crucial and specific roles in normal as well as pathological hematopoietic and in cancer cells.

Phosphoregulation of K+‐Cl− cotransporters during cell swelling: Novel insights
- BiologyJournal of cellular physiology
- 2018
It is found that WNK4 and PP1 regulate KCC4 activity as part of a common signaling module, but that they do not exert their effects through SPAK or carrier dephosphorylation, which implies that the response to cell swelling must also involve allosteric‐sensitive kinase‐dependent phosphoacceptor sites in KCC 4.

NH+4 is transported in a bumetanide-sensitive Na(+)-NH-4-Cl- mode, and both NH+4 and Rb+ (K+) are competitive inhibitors for the extracellular K+ binding site, however, the kinetics of Na( +)-K(-+)-2Cl- cotransport-mediated NH-4 transport differ from other K+ transport- mediated NH+ 4 pathways.
