The assembly and degradation of extracellular matrix (ECM) molecules are crucial processes during bone development. In this study, we show that ECM remodeling is a critical rate-limiting step in endochondral bone formation. Matrix metalloproteinase (MMP) 13 (collagenase 3) is poised to play a crucial role in bone formation and remodeling because of its expression both in terminal hypertrophic chondrocytes in the growth plate and in osteoblasts. Moreover, a mutation in the human MMP13 gene… Expand
Role of Matrix Metalloproteinase 13 in Both Endochondral and Intramembranous Ossification during Skeletal Regeneration
TLDR
It is demonstrated that M MP13 is involved in normal remodeling of bone and cartilage during adult skeletal repair, and that MMP13 may act directly in the initial stages of ECM degradation in these tissues prior to invasion of blood vessels and osteoclasts. Expand
Impaired remodeling phase of fracture repair in the absence of matrix metalloproteinase-2
TLDR
It is shown that MMPs play unique and distinct roles in regulating skeletal tissue deposition and remodeling during fracture repair and compared it with MMP9 and MMP13. Expand
Impaired bone fracture healing in matrix metalloproteinase-13 deficient mice.
TLDR
It is suggested that MMP-13 is crucial to the process of angiogenesis during healing of fracture, especially in the cartilage resorption process. Expand

TLDR
It is shown that administration of an EGFR‐specific small‐molecule inhibitor, gefitinib, into 1‐month‐old rats for 7 days produced profound defects in long bone growth plate cartilage characterized by epiphyseal growth plate thickening and massive accumulation of hypertrophic chondrocytes. Expand

TLDR
The results from in vitro and ex vivo studies suggest that collagen II matrikines stimulate angiogenesis in skeletal growth and development, revealing novel strategies for stimulating ang iogenesis in other contexts such as fracture healing and surgical applications. Expand
A Crucial Role for Matrix Metalloproteinase 2 in Osteocytic Canalicular Formation and Bone Metabolism*
TLDR
It is proposed that osteocytic network formation is a determinant of bone remodeling and mineralization and that MMP-2 plays a crucial role in forming and maintaining the osteocytics canalicular network. Expand
Galectin-3 is a downstream regulator of matrix metalloproteinase-9 function during endochondral bone formation.
TLDR
The disruption of growth plate homeostasis in Mmp-9 null mice links galectin-3 and MMP-9 in the regulation of the clearance of late chondrocytes through regulation of their terminal differentiation. Expand
ADAM17 Controls Endochondral Ossification by Regulating Terminal Differentiation of Chondrocytes
TLDR
The enlarged zone of hypertrophic chondrocytes in A17ΔCh mice resembles that described in mice with mutant EGFR signaling or lack of its ligand transforming growth factor α (TGFα), suggesting that ADAM17 regulates terminal differentiation of chondROcytes during endochondral ossification by activating the TGFα/EGFR signaling axis. Expand
Transforming growth factor alpha controls the transition from hypertrophic cartilage to bone during endochondral bone growth.
TLDR
Tgfa knockout mice experience a delay in bone development, specifically the conversion of hypertrophic cartilage to true bone, which appears to be mediated by a decrease in MMP13 and RANKL expression in hypertrophic chondrocytes and a resulting reduction in osteoclast recruitment. Expand
