Background: Modest affinity to carbohydrate targets limits the therapeutic efficacy of antibodies to tumor antigens, such as ganglioside GD2. Results: Mutagenesis and in silico modeling were utilized to enhance the affinity of monoclonal antibody hu3F8 to GD2. Conclusion: Alteration of electrostatic surface potential resulted in enhanced affinity and tumor cytotoxicity.
