Many growth factors exert their effects on cells by simulating intracellular kinases, which add phosphate groups to proteins. Two reports on p. 661 and p. 665 of this week's issue implicate the oncogenic kinase Akt in IGF-1-mediated neuronal survival (Dudek et al.) and show how lipids can regulate the activity of Akt (Franke et al.). In his Perspective, Hemmings discusses these results and several other recent reports about Akt and its unique mode of activation by phospholipids.
Signaling pathways and effector mechanisms pre-programmed cell death.
TLDR
This review discusses what is known about apoptotic pathways and its implication for normal cellular function and describes the characteristic morphological changes of apoptosis.
Role of Akt Signaling in Vascular Homeostasis and Angiogenesis
TLDR
In this review, Akt signaling in endothelial cells and its critical roles in the regulation of vascular homeostasis and angiogenesis will be discussed.
PKB and the mitochondria: AKTing on apoptosis.
TLDR
Mechanisms by which PKB mediates its anti-apoptotic activities in cells are summarized and recent developments in understanding mitochondrial dynamics and their role during apoptosis are surveyed.

This chapter will explore the role of survival factors in suppression of apoptosis, and illustrate how survival signals play a critical role in the survival of both normal and tumor cells. Survival
Membrane translocation and activation of the Akt kinase in growth factor-stimulated hematopoietic cells.
TLDR
It is demonstrated that overexpression of AKT2 protein is sufficient to transform NIH 3T3 cells, and that the oncogenic activity ofAKT2 is abolished by truncation of the 70amino acid proline-rich C-tail.
A novel conditional Akt ‘survival switch’ reversibly protects cells from apoptosis
TLDR
In Jurkat T cells stably expressing iAkt, CID-induced Akt activation rescued cells from apoptosis triggered by multiple apoptotic stimuli, including staurosporine, anti-Fas antibodies, PI3K inhibitors and the DNA damaging agent, etoposide.
Receptors that mediate cellular dependence
TLDR
Mechanistic studies of dependence receptors suggest that these receptors form ligand-dependent complexes that include specific caspases, analogous to the electrical integration system provided by dendritic arbors in the nervous system.
Akt/protein kinase B.
The phosphoinositide 3-kinases (PI3-K) and their downstream target kinase Akt play central roles in modulating fundamental cell functions, including survival, movement, and activation to produce
Genetic analysis of protein kinase B (AKT) in Drosophila
TLDR
A Drosophila PKB/AKT mutant (termed Dakt1) is isolated and characterized that exhibits ectopic apoptosis during embryogenesis as judged by induction of membrane blebbing, DNA fragmentation and macrophage infiltration, implicate Dakt 1 as a cell survival gene in Drosophile, consistent with cell protection studies in mammals.
Calcium promotes cell survival through CaM-K kinase activation of the protein-kinase-B pathway
TLDR
A Ca2+/calmodulin-dependent protein kinase kinase (CaM-KK) activates PKB directly, resulting in phosphorylation of BAD on serine residue 136 and the interaction of BAD with protein 14-3-3, which protects cells from apoptosis.

Requirement for phosphatidylinositol-3 kinase in the prevention of apoptosis by nerve growth factor.
TLDR
The ability of NGF to prevent apoptosis (programmed cell death) was inhibited by wortmannin or LY294002, two specific inhibitors of phosphatidylinositol (Pl)-3 kinase, and cell survival appears to be mediated by a Pl-3 Kinase signaling pathway distinct from the pathway that mediates differentiation.
A specific product of phosphatidylinositol 3-kinase directly activates the protein kinase Akt through its pleckstrin homology domain
TLDR
The findings show that a phospholipid product of PI 3-kinase can directly stimulate an enzyme known to be an important mediator of PI3,4P2-mediated activation, since a point mutation in the pleckstrin homology domain abrogated the response toPI3, 4P2.
GSK3: a SHAGGY frog story.
TLDR
Evidence is now accumulating that GSK3 plays a key role in the regulation of cell fate and differentiation in many eukaryotic species.
Regulating Cell Proliferation--As Easy as APC
TLDR
In his Perspective, Peifer discusses what the findings of this week's report on WINGLESS and WNT may mean for how APC participates in cell fate control.
Protein kinase B (c-Akt) in phosphatidylinositol-3-OH kinase signal transduction
TLDR
A role for PKB in PI(3)K-mediated signal transduction is suggested and a constructed Gag–PKB fusion protein, homologous to the v-akt oncogene, displays significantly increased ligand-independent kinase activity.
Direct Regulation of the Akt Proto-Oncogene Product by Phosphatidylinositol-3,4-bisphosphate
TLDR
Akt is apparently regulated by the direct interaction of PtdIns-3,4-P2 with the Akt PH domain, which is found to correlate with the amount of phosphatidylinositol-3.4-bisphosphate in vivo and in vitro and by the binding to other phosphoinositides, which revealed that they bound to the AkT PH domain with much lower affinity than did Ptd insurance and failed to increase Akt activity.
The protein kinase encoded by the Akt proto-oncogene is a target of the PDGF-activated phosphatidylinositol 3-kinase
TLDR
It is shown that Akt and the Akt-related kinase AKT2 are activated by PDGF, and it is suggested that the AkT PH domain may be a mediator of PI 3-kinase signaling.
Inhibition of glycogen synthase kinase-3 by insulin mediated by protein kinase B
TLDR
It is shown that agents which prevent the activation of both MAPKAP kinase-1 and p70S6k by insulin in vivo do not block the phosphorylation and inhibition of GSK3, and it is demonstrated that PKB is the product of the proto-oncogene protein kinase B (PKB, also known as Akt/RAC).
Akt, a Pleckstrin Homology Domain Containing Kinase, Is Activated Primarily by Phosphorylation*
TLDR
It is shown, in three cell types, that Akt does not require its pleckstrin homology domain to respond to either insulin or platelet-derived growth factor, and the primary mechanism of Akt activation is via protein phosphorylation.
Mouse p170 Is a Novel Phosphatidylinositol 3-Kinase Containing a C2 Domain*
TLDR
unique structural elements of mouse p170 include C-terminal sequences strikingly similar to the phosphoinositide-binding C2 domain of protein kinase C isoforms, synaptotagmins, and other proteins that define a new class of PI 3-kinase likely influenced by cellular regulators distinct from those acting upon p110- or VPS34-like PI3-kinases.
