AMP deaminase (AMPD) is essential for plant life, but the underlying mechanisms responsible for lethality caused by genetic and herbicide-based limitations in catalytic activity are unknown. Deaminoformycin (DF) is a synthetic modified nucleoside that is taken up by plant cells and 5′-phosphorylated into a potent transition state-type inhibitor of AMPD. Systemic exposure of Arabidopsis (Arabidopsis thaliana) seedlings to DF results in dose-dependent (150–450 nm) and time-dependent decreases in… Expand
Biochemical and physiological investigations on adenosine 5ʹ monophosphate deaminase from Plasmodium spp.

The combined results of these studies allow the identification of both short- and long-term adaptation of plant metabolism and development to direct changes in the adenylate status. Expand
Mechanism and structure based design of inhibitors of AMP and adenosine deaminase.
TLDR
It is proposed that the herbicidal activity of compound 16 is due to 5́-phosphorylation in planta to give an inhibitor of AMPD, which shows excellent selectivity for plant over human AMPD. Expand
PCR-based suppression subtractive hybridization analyses of enantioselective gene expression in root tips of wheat treated with optically active urea compounds
The optically active urea compounds R- and S-1-α-methylbenzyl-3-p-tolylurea (R- and S-MBTU) have qualitatively different effects on plant physiology, especially in Gramineae plants. To elucidate theExpand
The effect of adenosine monophosphate deaminase overexpression on the accumulation of umami-related metabolites in tomatoes
TLDR
This study highlights the changes in umami-related nucleotide and glutamate levels when the AMP deaminase gene was elevated in transgenic tomato and the effects on the levels of glutamate and GMP were unexpected. Expand
Characterization of a thermostable adenosine 5'-monophosphate deaminase gene in Streptomyces murinus.
TLDR
This study deals with the thermostable AMPD gene found from S. murinus, which is the best of the authors' knowledge, this is the fi rst bacterial AMPD to be identifi ed. Expand
Diversity in Guanosine 3′,5′-Bisdiphosphate (ppGpp) Sensitivity among Guanylate Kinases of Bacteria and Plants*
TLDR
It is shown that guanylate kinase (GK), a key enzyme in guanine nucleotide biosynthesis that catalyzes the conversion of GMP to GDP, is a target of regulation by ppGpp in chloroplasts of rice, pea, and Arabidopsis, and has uncovered diversity among bacterial GKs with regard to regulation byppGpp. Expand
Early Senescence in Older Leaves of Low Nitrate-Grown Atxdh1 Uncovers a Role for Purine Catabolism in N Supply1[OPEN]
TLDR
Results indicate that the absence of remobilized purine-degraded N from older leaves of Atxdh1 caused senescence symptoms, a result of higher chloroplastic protein degradation in older Leaves of low-nitrate-grown plants. Expand
Pre-mature senescence in the oldest leaves of low nitrate-grown Atxdh1 mutant uncovers a role for purine catabolism in plant nitrogen metabolism
TLDR
It is demonstrated that the absence of remobilized purine-degraded N from older leaves is the cause for senescence symptoms, a result of higher chloroplastic protein degradation in older leaves of nitrate starved Atxdh1 plants. Expand
Enhanced nucleotide analysis enables the quantification of deoxynucleotides in plants and algae revealing connections between nucleoside and deoxynucleoside metabolism.
TLDR
The potential of this new method to provide insight into plant NT metabolism is demonstrated, characterized by high sensitivity and robustness enabling the reproducible detection and absolute quantification of these metabolites even if they are of low abundance. Expand

5‐Aminoimidazole‐4‐carboxamide riboside activates nitrate reductase in darkened spinach and pea leaves
TLDR
The results suggest that a rise in cytosolic [5'-AMP] may be sufficient to activate NR in vivo, and are consistent with the notion that AICAR is converted to the monophosphorylated derivative, which accumulates in cells and acts as a structural analog of 5'-AMP. Expand
A Novel Superfamily of Transporters for Allantoin and Other Oxo Derivatives of Nitrogen Heterocyclic Compounds in Arabidopsis Article, publication date, and citation information can be found at www.plantcell.org/cgi/doi/10.1105/tpc.010458.
TLDR
It is shown that Arabidopsis plants are able to use allantoin as sole nitrogen source by functional complementation of a yeast mutant defective inAllantoin uptake, and AtUPS1 (Arabidopsis thaliana ureide permease 1), a novel superfamily of plant membrane proteins, is identified. Expand
Membrane Association, Mechanism of Action, and Structure of Arabidopsis Embryonic Factor 1 (FAC1)*
TLDR
An N-terminal transmembrane domain in Arabidopsis FAC1 is identified, subcellular fractionation is explored, and a 3.3-Å globular catalytic domain x-ray crystal structure with a bound herbicide-based transition state inhibitor is presented that provides the first glimpse of a complete AMPD active site. TLDR
5′-AMP-mimetic, 5-aminoimidazole-4-carboxamide ribonucleoside (AICAR), inhibits the fusicoccin-promoted proton extrusion in maize roots and the possible role of 5′-amp as a general regulator of 14-3-3 functions in the plant cell is discussed. Expand
Calcium activates erythrocyte AMP deaminase [isoform E (AMPD3)] through a protein-protein interaction between calmodulin and the N-terminal domain of the AMPD3 polypeptide.

Ca(2+)-CaM overcomes potent isoform E inhibitory mechanisms that function to maintain the total adenine nucleotide pool in mature erythrocytes, which are unable to synthesize AMP from IMP because of a developmental loss of adenylosuccinate synthetase. Expand

Studies using allopurinol, coformycin and tiazofurin, inhibitors of enzymes involved in purine metabolism, suggest that participation of AMP deaminase and xanthine oxidoreductase in the catabolism of adenine nucleotides in plant cells is suggested. Expand
EMBRYONIC FACTOR 1 encodes an AMP deaminase and is essential for the zygote to embryo transition in Arabidopsis.
