Neurometabolic diseases (NMDs) are typically caused by genetic abnormalities affecting enzyme functions, which in turn interfere with normal development and activity of the nervous system. Although the individual disorders are rare, NMDs are collectively relatively common and often lead to lifelong difficulties and high societal costs. Neuropsychiatric manifestations, including ADHD symptoms, are prominent in many NMDs, also when the primary biochemical defect originates in cells and tissues…

Impaired Amino Acid Transport at the Blood Brain Barrier Is a Cause of Autism Spectrum Disorder
TLDR
It is shown that the solute carrier transporter 7a5 (SLC7A5), a large neutral amino acid transporter localized at the blood brain barrier (BBB), has an essential role in maintaining normal levels of brain BCAAs and demonstrates that BCAA intracerebroventricular administration ameliorates abnormal behaviors in adult mutant mice.

Branched-chain α-ketoacid dehydrogenase deficiency (maple syrup urine disease): Treatment, biomarkers, and outcomes.
TLDR
There is a critical unmet need for safe and effective disease-modifying therapies for MSUD which can be implemented early in life and key insights into the design of new therapies based on gene replacement or editing are afforded.
ASL Metabolically Regulates Tyrosine Hydroxylase in the Nucleus Locus Coeruleus
TLDR
It is shown that ASL is prominently expressed in the nucleus locus coeruleus (LC), the central source of norepinephrine, and individuals with ASL deficiency are at risk for developing attention deficits.

TLDR
It is concluded that psychiatric manifestations in CTX follow a bimodal/bitemporal pattern, appearing early in the disease course in the form of a behavioral/personality disorder associated with learning difficulties or mental retardation, or manifesting in advanced disease in the setting of dementia as rich neuropsychiatric syndromes, such as frontal, orbitofrontal or frontotemporal syndrome of cortico-subcortical dementia.
Succinic semialdehyde dehydrogenase deficiency
TLDR
It was concluded that early urine organic acid analysis is essential for children presenting with mental retardation, neuropsychiatric disturbance or epilepsy of unknown etiology.
