PURPOSE Synthetic cathinones are designer analogs of the natural active principle of khat. Since their appearance on the black market in 2003, their popularity has increased annually, and they have become the most seized class of new psychoactive substances (NPS) reported to the UNODC Early Warning Advisory system. The constant introduction of newly synthesized molecules makes this issue difficult to monitor. The authors reviewed the most recent synthetic cathinone-related fatalities worldwide…
Synthetic Benzimidazole Opioids: The Emerging Health Challenge for European Drug Users
TLDR
With the broadening of the phenomenon toward Europe, in 2020, two-thirds of the global opioid-related fatal intoxications were attributed to NSO misuse, with the healthy-years life expectancy of opioid abusers decreased to 70% (6).
Abuse Potential of Cathinones in Humans: A Systematic Review
TLDR
Cathinone, mephedrone, methylone, and diethylpropion induce a range of desirable and reinforcing effects that may, to some extent, result in abuse potential.
Simple Analytical Strategy for Screening Three Synthetic Cathinones (α-PVT, α-PVP, and MDPV) in Oral Fluids
Synthetic cathinones are analogue compounds of the plant based stimulant cathinone. Its use, abuse, and related consumption complications have steadily increased in the last years. For this reason,
Evaluation of Cytotoxic and Mutagenic Effects of the Synthetic Cathinones Mexedrone, α-PVP and α-PHP
TLDR
Evaluated Mexedrone, α-PVP and α-PHP for mutagenicity evaluation for a complete assessment of the risk associated with synthetic cathinones exposure and demonstrated the mutagenic capacity of their metabolites.
Pyrrolidinyl Synthetic Cathinones α-PHP and 4F-α-PVP Metabolite Profiling Using Human Hepatocyte Incubations
TLDR
The metabolite profile of two pyrrolidinyl SCs, α-PHP and 4F-α-PVP, were characterized to identify optimal intake markers and an additional major metabolite that might be crucial for documenting exposure to α- PHP is identified.
A Quantitative LC–MS/MS Method for the Detection of 16 Synthetic Cathinones and 10 Metabolites and Its Application to Suspicious Clinical and Forensic Urine Samples
TLDR
This is the first report of NPS use in Saudi Arabia with respect to designer stimulant drugs and confirmatory urine analyses for suspicious stimulant use should extend beyond classical stimulants to cover a broad range of NPs and their metabolites in order to report any otherwise potentially undetected/new analyte.
Behavioural and neurochemical effects after repeated administration of N‐ethylpentylone (ephylone) in mice
TLDR
Investigating the neurochemical profile and the behavioural effects after both acute and repeated NEP exposure indicates that NEP consumption induces different neurological and neuropsychiatric disorders accompanied by changes in the monoaminergic system, posing a threat to public health.

Synthetic cathinones related fatalities: an update.
TLDR
Use of synthetic cathinones still represents a serious public health issue and Analytical methodologies for the identification of parent compounds and eventual metabolites both in ante-mortem and post-mortem cases need to be developed and validated.
The synthetic cathinone α-pyrrolidinovalerophenone (α-PVP): pharmacokinetic and pharmacodynamic clinical and forensic aspects
TLDR
There is a limited knowledge regarding the short- and long-term effects of α-PVP and metabolites, and pharmacogenetic influence, hence further clinical and forensic toxicological studies are required.
Fatal case of poisoning with a new cathinone derivative: α-propylaminopentiophenone (N-PP)
TLDR
This case is reported of the purposeful intake of a high dose of powder containing a novel cathinone derivative, α-propylaminopentiophenone, which resulted in the deadly poisoning of a woman.
n‐Ethyl Pentylone‐Related Deaths in Alabama
TLDR
Four fatalities involving NEP from Alabama in 2017 are presented to enhance the forensic literature regarding specific NPS like NEP and provide contextual reference for professionals considering the significance of NEP in toxicological interpretation.

First metabolic profile of PV8, a novel synthetic cathinone, in human hepatocytes and urine by high-resolution mass spectrometry
TLDR
To determine PV8’s metabolic stability in human liver microsome incubation and its metabolism following human hepatocyte incubation, and to focus reference standard companies’ synthetic efforts to provide commercially available standards needed for PV8 biological specimen testing, data are analyzed.
