OBJECTIVES A randomized, placebo-controlled, double-blind trial of the relative effectiveness of glucosamine sulphate and placebo in managing pain in osteoarthritis (OA) of the knee. METHODS Eighty patients with OA of the knee were recruited from a rheumatology out-patient clinic and received either glucosamine sulphate 1500 mg daily for 6 months or dummy placebo. The primary outcome measure was patients' global assessment of pain in the affected knee. RESULTS Area under the curve analysis…

Randomised, Double-Blind, Parallel, Placebo-Controlled Study of Oral Glucosamine, Methylsulfonylmethane and their Combination in Osteoarthritis
TLDR
The combination of MSM with Glu provides better and more rapid improvement in patients with osteoarthritis and shows better efficacy in reducing pain and swelling and in improving the functional ability of joints than the individual agents.
Short report: ibuprofen versus glucosamine sulfate. Treating osteoarthritis pain.
TLDR
Evaluating the efficacy and side effect profile of glucosamine sulfate in a 12-week double-blind pilot study aimed to help resolve the ongoing controversy by further evaluating the effects of the drug in osteoarthritis patients.
Intraarticular injection of anakinra in osteoarthritis of the knee: a multicenter, randomized, double-blind, placebo-controlled study.
TLDR
Anakinra was well tolerated as a single 50-mg or 150-mg intraarticular injection in patients with OA of the knee, however, anakinRA was not associated with improvements in OA symptoms compared with placebo.
Subgroup analyses of the effectiveness of oral glucosamine for knee and hip osteoarthritis: a systematic review and individual patient data meta-analysis from the OA trial bank
TLDR
There is no goodEvidence to support the use of glucosamine for hip or knee OA and an absence of evidence to support specific consideration of glucOSamine for any clinically relevant OA subgroup according to baseline pain severity, BMI, sex, structural abnormalities or presence of inflammation.
A Double-Blind Randomized Placebo Controlled Parallel Group Study Evaluating the Effects of Ibuprofen and Glucosamine Sulfate on Exercise Induced Muscle Soreness
TLDR
Glucosamine sulphate facilitates Muscle tenderness whereas systemic administration of ibuprofen is not capable of inhibiting experimentally induced muscle tenderness/soreness.
Glucosamine for pain in osteoarthritis: why do trial results differ?
TLDR
Heterogeneity among trials of glucosamine is larger than would be expected by chance, and potential explanations include different glucosamines preparations, inadequate allocation concealment, and industry bias.
Glucosamine Long-Term Treatment and the Progression of Knee Osteoarthritis: Systematic Review of Randomized Controlled Trials
TLDR
The available evidence suggests that glucosamine sulfate may be effective and safe in delaying the progression and improving the symptoms of knee OA.
Glucosamine therapy for treating osteoarthritis.
TLDR
The results are not uniformly positive, and the reasons for this remain unexplained, but glucosamine was superior to placebo in the treatment of pain and functional impairment resulting from symptomatic OA.

Evaluation of benefit of glucosamine and chondroitin preparations for OA symptoms using meta-analysis combined with systematic quality assessment of clinical trials of these preparations in knee and/or hip OA suggests some degree of efficacy appears probable for these preparations.
