RATIONALE Conventionally, myofibrillar protein synthesis is measured over time periods of hours. In clinical studies, interventions occur over weeks. Functional measures over such periods may be more representative. We aimed to develop a novel method to determine myofibrillar protein fractional synthetic rate (FSR) to estimate habitual rates, while avoiding intravenous tracer infusions. METHODS Four healthy males were given 100 g water enriched to 70 Atom % with (2)H2O as a single oral bolus…
Habitual Myofibrillar Protein Synthesis Is Normal in Patients with Upper GI Cancer Cachexia
TLDR
The finding implies a small increase in muscle breakdown may account for muscle wasting, and there was no evidence of suppression of myofibrillar protein synthesis in patients with cancer cachexia.
Measurement of protein digestibility in humans by a dual-tracer method
TLDR
An innovative, minimally invasive “dual-stable-isotope” method was developed to measure protein digestibility, in which the ingestion of an intrinsically 2H- labeled test protein along with a 13C-labeled standard protein of known digestibility allows for an accurate measure of digestion and absorption of the intrinsically labeled protein.
A validation of the application of D(2)O stable isotope tracer techniques for monitoring day-to-day changes in muscle protein subfraction synthesis in humans.
TLDR
D2O has unrivaled utility to quantify day-to-day MPS in humans and inform on short-term changes in anabolism and presumably catabolism alike.
Integrated Myofibrillar Protein Synthesis in Recovery From Unaccustomed and Accustomed Resistance Exercise With and Without Multi-ingredient Supplementation in Overweight Older Men
TLDR
The finding that supplementation did not alter 0–48 h MyoPS following 12 weeks of training suggests a possible adaptive response to longer-term increased protein intake and warrants further investigation.
Daily resistance-type exercise stimulates muscle protein synthesis in vivo in young men.
TLDR
It is demonstrated that daily resistance-type exercise stimulates muscle protein synthesis rates in vivo in humans over multiple days and that saliva deuterium enrichment serves as an appropriate and practical choice of precursor.
Whey protein but not collagen peptides stimulate acute and longer-term muscle protein synthesis with and without resistance exercise in healthy older women: a randomized controlled trial.
BACKGROUND Aging appears to attenuate the response of skeletal muscle protein synthesis (MPS) to anabolic stimuli such as protein ingestion (and the ensuing hyperaminoacidemia) and resistance
The application of stable‐isotope tracers to study human musculoskeletal protein turnover: a tale of bag filling and bag enlargement
TLDR
Over a 30 year period, the way in which amino acids and insulin mediate the anabolic effect of a meal has been elaborated in great detail confirming the original concepts of bag filling within the muscle endomysial “bag”, which is limited by the “ bag” size unless bag enlargement occurs requiring new collagen synthesis.
HabitualMyo fi brillarProteinSynthesis IsNormal in Patients with Upper GI Cancer Cachexia
TLDR
The finding implies a small increase in muscle breakdown may account for muscle wasting, and there was no evidence of suppression of myofibrillar protein synthesis in patients with cancer cachexia.
Assessing the regulation of skeletal muscle plasticity in response to protein ingestion and resistance exercise: recent developments
TLDR
Novel insights into cellular signaling processes may help delineate the molecular mechanisms that govern skeletal muscle plasticity in response to exercise and feeding and test the long-term efficacy of interventions to enhance skeletal muscle protein reconditioning and hypertrophy.
Day-to-Day Changes in Muscle Protein Synthesis in Recovery From Resistance, Aerobic, and High-Intensity Interval Exercise in Older Men.
TLDR
It is shown that in older men, changes in muscle protein synthesis in response to certain exercises are long lasting and that HIIT significantly increases myofibrillar and sarcoplasmic fractional synthetic rate in this population.

Human muscle protein turnover--why is it so variable?
TLDR
The results from studies that report the protein fractional synthesis rate (FSR) in the vastus lateralis in healthy, nonobese, untrained adults ≤50 yr of age in the postabsorptive state at rest by using the primed, constant tracer amino acid infusion method are analyzed.

TLDR
This work has sought to improve the situation by use of an alternative derivative that overcomes this problem whilst also streamlining sample preparation, and analysed albumin-bound Phe as ECTFE-phe with improved precision, independent of the isotopomer used.
The application of 2H2O to measure skeletal muscle protein synthesis
TLDR
The framework provided in this review should assist muscle biologists in designing experiments that advance the understanding of conditions in which anabolism is altered (e.g., exercise, feeding, growth, debilitating and metabolic pathologies).
Measurement of protein turnover rates by heavy water labeling of nonessential amino acids.
Measurement of liver collagen synthesis by heavy water labeling: effects of profibrotic toxicants and antifibrotic interventions.
TLDR
Changes in isotopically measured collagen synthesis correlated with, but were more sensitive and reproducible than, standard histological staining (trichrome) for fibrosis, thereby providing a relatively high-throughput in vivo strategy for rapidly measuring profibrotic activities of suspected hepatotoxicants and antifibrotics activities of drug candidates.
Quantifying rates of protein synthesis in humans by use of 2H2O: application to patients with end-stage renal disease.
TLDR
Estimates are in subjects with renal disease, but the method should be applicable to estimates of protein synthetic rates in normal subjects and in other pathological states, and an integrated measure over a long period of albumin's synthetic rate can be estimated from both the rise in enrichment of alanine from the protein during (2)H(2)O ingestion and fall on (2]H( 2)O withdrawal, while the subject's living routine is uninterrupted.
Myofibrillar and collagen protein synthesis in human skeletal muscle in young men after maximal shortening and lengthening contractions.
TLDR
A more rapid rise in MPS after maximal LC could translate into greater protein accretion and muscle hypertrophy during chronic resistance training utilizing maximal LC.

TLDR
The application of new stable isotope methods using heavy water to animals other than rodents will undoubtedly enhance the understanding of adipose tissue biology and could lead to new methods for improving animal production.
A streamlined approach to the analysis of volatile fatty acids and its application to the measurement of whole-body flux.
TLDR
A streamlined and robust method will permit rapid and accurate measurement of VFA flux using [(2)H]- and/or [(13)C]-labelled VFAs as tracers, and Measurements of the contribution of colonic VFA production to whole-body V FA flux are now possible.
