Corynebacterineae are characterized by the presence of long-chain lipids, notably mycolic acids (α-alkyl, β-hydroxy fatty acids), the structures of which are genus-specific. Mycolic acids from two environmental strains, Amycolicicoccus subflavus and Hoyosella altamirensis, were isolated and their structures were established using a combination of mass spectrometry analysis, (1)H-NMR spectroscopy and chemical degradations. The C(2)-C(3) cleavage of these C(30)-C(36) acids led to the formation of… Expand
Structural elucidation and genomic scrutiny of the C60-C100 mycolic acids of Segniliparus rotundus.
TLDR
Examination of the Segniliparus rotundus genome indicated two obvious differences in genes encoding the elongation fatty acid (FAS-II) enzymes involved in the biosynthesis of mycolic acids: the organization of 3-ketoacyl-ACP synthases (KasA and KasB), on one hand, and the presence of two copies of the hadB gene encoding the catalytic domain of the latter enzyme type, on the other. Phylogeny Trumps Chemotaxonomy: A Case Study Involving Turicella otitidis
TLDR
Polyphasic phylogenetic analyses using 16S rRNA, bacterial core genes and genes responsible for synthesizing menaquinones unequivocally indicate that Turicella is a true member of the genus Corynebacterium. Expand
Reclassification of Amycolicicoccus subflavus as Hoyosella subflava comb. nov. and emended descriptions of the genus Hoyosella and Hoyosella altamirensis.
16S rRNA gene sequences of two type strains belonging to different genera within the suborder Corynebacterineae, namely Hoyosella altamirensis and Amycolicicoccus subflavus, show a similarity of 99.8Expand
HadD, a novel fatty acid synthase type II protein, is essential for alpha- and epoxy-mycolic acid biosynthesis and mycobacterial fitness
TLDR
The change in the envelope composition induced by hadD inactivation strongly altered the bacterial fitness and capacities to aggregate, assemble into colonies or biofilms and spread by sliding motility, and conferred a hypersensitivity to the firstline antimycobacterial drug rifampicin. Expand
Mycolic acids: structures, biosynthesis, and beyond.
TLDR
Progress in deciphering the mycolic acid biosynthetic pathway is described and the functional and key biological roles of these molecules are discussed, providing a historical perspective in this dynamic area. Expand
Cell Walls and Membranes of Actinobacteria.
TLDR
This chapter describes the main structural and biosynthetic features of major components found in the actinobacterial cell envelopes and highlights the key differences between them. Expand
Genome-based exploration of the specialized metabolic capacities of the genus Rhodococcus
TLDR
The genus Rhodococcus provides an interesting target for natural product discovery, in view of its large and mostly uncharacterized biosynthetic repertoire, its relatively fast growth and the availability of effective genetic tools for its genomic modification. Expand
Current Perspectives on Mycobacterium farcinogenes and Mycobacterium senegalense, the Causal Agents of Bovine Farcy

TLDR
Molecular analyses confirmed that M. farcinogenes and M. senegalense fall in a subclade together with M. houstonense andM. Expand
The changes in mycolic acid structures caused by hadC mutation have a dramatic effect on the virulence of Mycobacterium tuberculosis
TLDR
It is shown here that mutation or deletion of hadC affects the biosynthesis of oxygenated mycolic acids, substantially reducing their production level, and causes the loss of atypical extra‐long mycoli acids, demonstrating the involvement of HadBC in the late elongation steps of my colic acid biosynthesis. Expand
Bacterial natural products: Prediction, regulation and characterization of biosynthetic gene clusters in Actinobacteria
TLDR
Important insights are given in the (regulation of) secondary metabolism in S. clavuligerus ATCC 27064 and members of the Rhodococcus genus, revealing an impressive diversity of (novel) secondary metabolite gene clusters. Expand

Mycolic acid analysis in Nocardia species. The mycolic acid compositions of Nocardia asteroides, N. farcinica, and N. nova.
TLDR
The profiles of the mycolic acid molecular species were used as models to illustrate the difference in the structures of each species, even in the case of N. asteroides or N. nova. Expand
Tetraenoic and pentaenoic mycolic acids from Mycobacterium thamnopheos. Structure, taxonomic and biosynthetic implications.
TLDR
Results suggested that M. thamnopheos might be more related to the Aurantiaca taxon than to mycobacteria and Nocardia, and the relative configuration of both mycolates was found to be threo, like that established for all mycolate studied so far. Expand
Mycolic acids from Rhodococcus, Gordonia, and Dietzia.
TLDR
The genus Dietzia was found to have its own characteristic constitution of mycolic acid molecular species, characterized by an almost equal amount of constituents of even- and odd-numbered carbon chains, whereas the major components of my colic acids in all other strains had even- numbered carbon chains. TLDR
This study revealed that the mycolic acids isolated from pathogenic Rhodococcus equi 103 contained a series of homologous ions having C(30) to C(50) chain with 0-2 double bonds, which enabled structural assignment of the myColic acids, including the lengths of the meromycolate chain and the α-branch. In silico analysis of various mycobacterial genomes, results indicate that Pks13 is the condensase, a promising target for the development of new antimicrobial drugs against Corynebacterineae. Expand
The biosynthesis of mycolic acids by Mycobacteria: current and alternative hypotheses.
TLDR
The hypothetical coexistence of two pathways for mycolate biosynthesis is discussed, where some methyl or methylene substituents or oxygenated functions are added to the double bonds of an unsaturated precursor, whereas in the second hypothesis, the methylations could help in the building of very long aliphatic chains, and determine the location of double bonds or ramifications.
