During our on-going attempts to develop a new skin-whitening agent, we identified a novel candidate compound KHG22394, a 2-imino-1,3-thiazoline derivative. Our data show that KHG22394 significantly inhibits melanin production in a dose-dependent manner, but that it does not directly inhibit tyrosinase, the rate limiting melanogenic enzyme. It has been reported that the activation of extracellular signal-regulated kinase (ERK) reduces melanin synthesis by downregulating microphthalmia-associated…

Figures and Tables from this paper

A novel adamantyl benzylbenzamide derivative, AP736, inhibits melanogenesis in B16F10 mouse melanoma cells via glycogen synthase kinase 3β phosphorylation.
TLDR
The data suggest that AP736 exerts hypopigmentary effects through the downregulation of tyrosinase via GSK3β phosphorylation, a pathway of melanogenesis involving the effects of AP736 on the glycogen synthesis kinase 3β (GSK3β) pathway.
A novel psoralen derivative-MPFC enhances melanogenesis via activation of p38 MAPK and PKA signaling pathways in B16 cells.
TLDR
Results indicated that MPFC-mediated activation of the p38 MAPK and the protein kinase A (PKA) pathway may shed light on a novel approach for an effective therapy for vitiligo.
Hypo-pigmenting effect of sesquiterpenes from Inula britannica in B16 melanoma cells
TLDR
Results demonstrated that compound 1, a major sesquiterpene from the flowers of I. britannica, exhibited anti-melanogenic activity by suppression of tyrosinase expression via ERK and Akt signaling, suggesting that these compounds may act as potent natural skin-lightening agents.
Di (Isoquinolin-1-Yl) Sulfane (DIQS) Inhibits Melaninogenesis by Modulating PKA/CREB and MAPK Signaling Pathways
TLDR
The results suggest that DIQS attenuates hyperpigmentation via inhibition of the cAMP/PKA/CREB/MITF/tyrosinase axis and MAPK pathways and the potential ofDIQS as a small-molecule agent for skin-whitening cosmetics and the treatment of hyperpIGmentation disorders without biological toxicity is suggested.
Ssanghwa-tang, an oriental herbal cocktail, exerts anti-melanogenic activity by suppression of the p38 MAPK and PKA signaling pathways in B16F10 cells
TLDR
Results indicate that SHT may be useful as herbal medicine for treating hyperpigmentation and cosmetics as a skin-whitening agent and as an herbal cocktail showed synergistic anti-melanogenic effect compared with that of each single constituent herb.
[6]-Shogaol inhibits melanogenesis in B16 mouse melanoma cells through activation of the ERK pathway
TLDR
It is demonstrated that [6]-shogaol inhibits melanogenesis in B16F10 mouse melanoma cells via activating the ERK pathway.

Sphingosine-1-phosphate decreases melanin synthesis via sustained ERK activation and subsequent MITF degradation
TLDR
It is suggested that S1P reduces melanin synthesis by ERK activation, MITF phosphorylation and degradation, and by the subsequent downregulation of tyrosinase and TRP-1 production.
Inhibition of the Phosphatidylinositol 3-Kinase/p70S6-Kinase Pathway Induces B16 Melanoma Cell Differentiation*
TLDR
Evidence is provided which suggests that cAMP-induced melanogenesis and dendricity are, at least partially, mediated by the cAMP inhibition of the phosphatidylinositol 3-kinase/p70S6-kinases signaling pathway.
Inhibition of the Mitogen-activated Protein Kinase Pathway Triggers B16 Melanoma Cell Differentiation*
TLDR
Results demonstrate that the MAP kinase pathway activation is not required for cAMP-induced melanogenesis, and the inhibition of this pathway induces B16 melanoma cell differentiation, while a sustained activation impairs the melanogenic effect of cAMP -elevating agents.
Development of an in vitro primary screen for skin depigmentation and antimelanoma agents.
TLDR
All of the compounds identified as active in this primary screen were cytotoxic or cytostatic to melanocytes, except for the methyl ester of gentisic acid, which uniquely inhibited the de novo synthesis of melanin without cytotoxicity.
Alpha-melanocyte stimulating hormone and its analogue Nle4DPhe7 alpha-MSH affect morphology, tyrosinase activity and melanogenesis in cultured human melanocytes.
TLDR
The preliminary EC50 values for the actions of the MSH peptides suggest that they may be equipotent in their actions on human melanocytes, and it is demonstrated that the common melanocyte mitogens 12-O-tetradecanoyl phorbol-13-acetate (TPA) and cholera toxin affect basal melanogenesis and modulate the effects of theMSH peptide.
The Microphthalmia gene product interacts with the retinoblastoma protein in vitro and is a target for deregulation of melanocyte-specific transcription.
TLDR
The results obtained show that the adenovirus E1A 13S, but not the 12S, product can transform the highly pigmented and TPA-dependent melanocyte cell line melan-a, and that the mi gene appears to play a crucial role in melanocyte-specific gene expression.
Cyclic AMP a key messenger in the regulation of skin pigmentation.
TLDR
It appears that melanogenic gene expression is controlled by a complex network of regulation involving other transcription factors such as Brn2, TBX2, PAX3 and SOX10, which would finally allow a fine tuning of melanocyte differentiation leading to melanin synthesis.
Tyrosinase related protein 1 (TRP1) functions as a DHICA oxidase in melanin biosynthesis.
TLDR
The data demonstrate that the specific melanogenic function of TRP1 is the oxidation of 5,6‐dihydroxyindole‐2‐carboxylic acid (DHICA) to a carboxylated indole‐quinone at a down‐stream point in the melanin biosynthetic pathway.
