}An IgA monoclonal antibody (MUM/F1-1/copenhageni) was produced from a mouse immunised with Leptospira interrogans serovar copenhageni. The antibody showed partial serogroup specificity by agglutination and by reaction in enzyme immunoassay, and opsonised homologous leptospires for phagocytosis by cultured mouse macrophages. Immunodiffusion and Western-blotting experiments indicated that MUM/F1-1/copenhageni reacted with a carbohydrate determinant in the leptospiral lipopolysaccharide. Daily…

Immunocytochemistry showed that an EDTA extract determinant recognized by monoclonal antibody 47B4D6, is localized under the outer envelope of serovar icterohaemorrhagiae, and was not digested by pronase, suggesting that the determinant is of carbohydrate nature.

Serum samples obtained from patients hospitalized in Barbados with severe leptospirosis were tested by the microscopic agglutination test (MAT), enzyme immunoassay (EIA) and immunoblotting withLeptospires that had been isolated from these patients, revealing reactions with a number of bands that corresponded with outer envelope components.

A sustained IgM response potentially associated with chronic leptospiral renal infection is observed in mice infected with L. interrogans, and it is demonstrated that mice pre-infected with the attenuated M895 Manilae LPS mutant or heat-killed bacterin were completely protected against renal colonization.

The type-specific main (TM) antigen was extracted from the leptospiral cells of strain Kyoto of the Hebdomadis group by 90% phenol and was purified mainly by ethanol precipitation and generally resembled that of gram-negative bacterial endotoxin, and this fact suggests that the antigen is composed mainly of lipopolysaccharides.
