The use of chemotherapy to treat cancer began at the start of the 20th century with attempts to narrow the universe of chemicals that might affect the disease by developing methods to screen chemicals using transplantable tumors in rodents. It was, however, four World War II-related programs, and the effects of drugs that evolved from them, that provided the impetus to establish in 1955 the national drug development effort known as the Cancer Chemotherapy National Service Center. The ability of…

Chemotherapy over the Years
TLDR
The article highlights the major milestones in chemotherapy through the years, including the first fruitful application of a multifaceted tactic to treat cancer in children in the 1960s and the early 1970s.
A CONCISE HISTORY ON CHEMOTHERAPY
TLDR
This essay aims to be a contribution to the study of chemotherapy, namely in the description of its history, indicating the main types of and treatments in chemotherapy.
Major Contributions towards Finding a Cure for Cancer through Chemotherapy: A Historical Review
TLDR
In this review an attempt is made to discuss the major breakthroughs that have shaped the course of cancer chemotherapy, helping to decrease the mortality as well as lessen the suffering of patients.
Exploiting Cancer’s Tactics to Make Cancer a Manageable Chronic Disease
TLDR
The tactics employed in cancer evolution; namely, chromosomal instability (CIN), intra-tumoral heterogeneity (ITH), and cancer-specific metabolism are analyzed, which govern the resistance to current cancer therapeutics.
Anti-cancer Drugs: Discovery, Development and Therapy
TLDR
The impact of the molecular understanding of cancer on modern drug discovery, drug development and cancer therapy will be explored to transform cancer treatments from cytotoxic regimens towards the rapidly dividing cells into personalized targeted therapies.
Drug Combination in Cancer Treatment—From Cocktails to Conjugated Combinations
TLDR
The evolution of chemotherapy from its beginning as a single-agent based therapy to poly-drug treatment that involves targeted drugs, and the concept of drug-conjugation based treatment as a strategy for further optimization of treatment regimes is outlined.
RECENT ADVANCES IN THE DEVELOPMENT OF ANTINEOPLASTIC AGENT
TLDR
The review highlights the recent development of antibody-drug conjugates and other immunotoxins, which are capable of delivering highly cytotoxic agents previously deemed too toxic to elicit therapeutic benefit preferentially to neoplastic cells.
Modern Approaches to Testing Drug Sensitivity of Patients’ Tumors (Review)
TLDR
The present review considers the main approaches to drug sensitivity assessment of patients’ tumors: molecular genetic profiling of tumor cells, and direct efficiency testing of the drugs on tumor cells isolated from surgical or biopsy material.
Anticancer Drug Development Unique Aspects of Pharmaceutical Development
TLDR
This chapter will highlight the unique aspects of anticancer drug discovery and development, which differ significantly from the drug development process for any other indication.
Advancing Therapies for Cancer—From Mustard Gas to CAR T
TLDR
Therapies with improved specificity, localization, and efficacy are redefining cancer treatment and the march towards personalized medicine in the treatment of human cancer is reviewed.

Combination versus single agent chemotherapy: A review of the basis for selection of drug treatment of cancer
TLDR
Now that the chemotherapeutic tools are sharpened, their use in combinations with other modalities in the previously unfamiliar setting of the patient with early stages of the disease promises to lead to an even more exciting chapter in clinical cancer research in the next decade.
Approaches to Tumor Chemotherapy
TLDR
The doyen of American cancer research, W. H. Woglom, believes that hormone therapy has given the first hopes of ultimate success in the untiring attack on malignant disease by chemotherapy.
Effects of combined drug therapy on metastatic cancer of the testis.
TLDR
Tumor regression was associated with a fall of urinary chorionic gonadotropin titer and side-effects occurred but proved reversible, and they were accepted as hazards of the treatment because of the striking improvement obtained in some cases.

TLDR
The modern era of chemotherapy developed at Yale University Medical School during World War II, a fact that has been generally unrecognized until recently.
Intensive combination chemotherapy and X-irradiation in Hodgkin's disease.
TLDR
It appears that the incidence of complete remissions can be increased over that obtained with a single agent and the duration of complete unmaintained remission in those patients with advanced disease is provocative even though the group is small.
Concepts for systemic treatment of micrometastases
TLDR
Since first‐order cell kill kinetics characterizes effective drug kill of tumor cells and the best currently known anticancer drugs have variable but generally limited tumor cell kill potential, drug treatment of likely micrometastases should be started as soon after the end of likely noncurative radiologic or surgical treatment as possible.
Some observations on the effect of folic acid antagonists on acute leukemia and other forms of incurable cancer.
TLDR
No evidence has been presented which would justify the use of the word "cure" of acute leukemia, and two of the most pressing problems demanding solution are concerned with the nature, the prevention, and the treatment of toxic changes, including hemorrhage, produced by these folic acid antagonists.
Actinomycin D in childhood cancer; a preliminary report.
TLDR
Actinomycin D was administered intravenously to 13 children with inoperable cancer and two experienced regression of tumor, of whom one had Wilms9 tumor and the other rhabdomyosarcoma, and the changes in the skin associated with x-radiation appeared to become exaggerated after therapy with Actinomycus D.
The effect of chemotherapy on the growth characteristics and cellular kinetics of leukemia L1210.
TLDR
Research on the regrowth of the L1210 cells exposed to 1,3-bis(2-chloroethyl)-1-nitrosourea but not sustaining a lethal biochemical lesion demonstrated a normal duration of DNA synthesis and generation time and an alteration of the proliferating pool size.
Testicular cancer as a model for a curable neoplasm: The Richard and Hinda Rosenthal Foundation Award Lecture.
TLDR
This third-generation study tested the hypothesis of whether maintenance vinblastine was necessary to ensure optimal cure rates in disseminated testicular cancer and demonstrated that cure in a far-advanced cancer could be achieved with only 12 weeks of therapy (remission induction) because the relapse rate in such patients was only 7%.
