We have developed a miniaturized and multiplexed solution assay for the measurement of protease activity in complex samples. This technology can accelerate research in functional proteomics and enable biologists to carry out multiplexed protease inhibitor screens on a large scale. The assay readout is based on Illumina's universal Sentrix BeadArrays. The peptide sequences that serve as protease substrates are conjugated to oligonucleotide sequences complementary to the oligo tags on randomly…

Deciphering Enzyme Function Using Peptide Arrays
TLDR
Focus of this review is on assay principles for measuring activities of kinases, phosphatases or proteases and on substrate identification/optimisation for kinases and high-density peptide microarrays for the simultaneous profiling of kinase activities in complex biological samples.
Endoprotease profiling with double-tagged peptide substrates: a new diagnostic approach in oncology.
TLDR
A mass spectrometry (MS)-based functional proteomic profiling approach that tracks degradation of artificial endoprotease substrates in serum specimens and the introduction of a few structural key elements into synthetic reporter peptides increases the diagnostic sensitivity for MS-based protease profiling of serum specimens.
Peptide arrays for enzyme profiling.
TLDR
A comprehensive overview regarding enzyme profiling using peptide microarrays is presented with special focus on assay principles and reduces set-up time for HTS assays and allows the identification of downstream targets.
Peptide microarrays as diagnostic tool for cancer associated protease profiling
TLDR
This PhD thesis dealt with the development of a high-throughput screening assay platform based on in-situ synthesized peptide microarrays, using a biotin-streptavidin system to minimize the peptide label to a smaller tag, which allowed greater sequence variability and avoided false positive cleavage, when using a proteinogenic tag sequence.
Substrate optimization and clinical validation of reporter peptides for MS-based protease profiling in serum specimens: a new approach for diagnosis of malignant disease.
TLDR
The results demonstrate that RPs for MS-based protease profiling can systematically be optimized with a positional scanning synthetic combinatorial library (PS-SCL) approach and the combination of different RPs can additionally increase the classification accuracy of functional protease profiles, and this in turn could lead to improved diagnosis, monitoring and prognosis of malignant disease.
A Novel Continuous Assay for the Deacylase Sirtuin 5 and Other Deacetylases.
TLDR
An improved continuous sirtuins 5 assay based on the coupling of the sirtuin reaction to a proteolytic cleavage using internally fluorescence-quenched substrates is described, which now enables more efficient development of sIRTuin targeting drugs.
Functional protease profiling for diagnosis of malignant disease
TLDR
The present article summarizes and discusses the current status of proteomic techniques to identify tumour‐specific protease‐substrate interactions for functional protease profiling and focuses on the potential diagnostic use of the respective approaches.
Substrate Cleavage Analysis of Furin and Related Proprotein Convertases
TLDR
The results suggest that pathogens, including anthrax PA83 and the avian influenza A H5N1 (bird flu) hemagglutinin precursor, evolved to be as sensitive to PC proteolysis as the most sensitive normal human proteins.
Decoration of trastuzumab with short oligonucleotides: synthesis and detailed characterization.
TLDR
A simple, fast and cost-effective methodology to conjugate trastuzumab with 22-mer 5' thiol-modified oligonucleotides using a bifunctional crosslinker is proposed.
Funktionelles Protease-Profiling als neue onkologische Diagnosestrategie
TLDR
Tumour-associated protease activity in serum specimens of cancer patients can be monitored using synthetic reporter peptides and this hopefully might facilitate diagnosis and improve prognosis of malignant disease.

Evaluation of Different Chemical Strategies for Conjugation of Oligonucleotides to Peptides
TLDR
Novel assays for high-throughput detection of one or many kinases or proteases, each covalently linked to an oligonucleotide tag, and validated selected method during the successful generation of thousands oligon nucleotide-peptide conjugates are developed.
