Transporter associated with antigen processing (TAP), a member of the ATP-binding cassette transporter superfamily, is composed of two integral membrane proteins, TAP-1 and TAP-2. Each subunit has a C-terminal nucleotide-binding domain that binds and hydrolyzes ATP to energize peptide translocation across the endoplasmic reticulum membrane. A motif comprising the sequence LSGGQ (called the signature motif) and the amino acid that is immediately C-terminal to this motif are highly conserved in…

Catalytic Site Modifications of TAP1 and TAP2 and Their Functional Consequences*
TLDR
It is found that alterations at Glu632 and His661 of TAP2 significantly reduced peptide translocation and/or TAP-induced major histocompatibility complex class I surface expression.
Functionally important interactions between the nucleotide-binding domains of an antigenic peptide transporter.
TLDR
It is demonstrated biochemically that there are two electrostatic interactions across the interface between the two TAP NBDs and that these interactions are important for peptide transport, suggesting that the interactions function at alternative stages in the transport cycle.
Association of TAP1 and TAP2 gene polymorphisms with hematological malignancies.
TLDR
It could be concluded that the TAP2-665 GG genotype might be a risk factor for all types of hematological malignancies included in this study.
The mechanism of ABC transporters: general lessons from structural and functional studies of an antigenic peptide transporter
TLDR
A potential mechanism for the TAP protein is outlined with supporting evidence from bacterial transporter structures and the notion that ABC transporters in general have adapted around a universal transport mechanism is supported.
The genotype of the transporter associated with antigen processing gene affects susceptibility to colorectal cancer in Japanese
TLDR
The genotype of the TAP1 gene is associated with susceptibility to CRC, and a significant trend in the rate of cases was observed with an increasing number of mutated alleles.
HLA-B44 Polymorphisms at Position 116 of the Heavy Chain Influence TAP Complex Binding via an Effect on Peptide Occupancy1
TLDR
These studies suggest that position 116 HLA polymorphisms influence peptide occupancy, which in turn determines binding to tapasin and TAP.
Genetic variation of antigen processing machinery components and association with cervical carcinoma
TLDR
Combined genetic variation in the TAP2, LMP7, and ERAP1 genes is associated with increased cervical carcinoma risk, and SNP genotypes and haplotypes are modeled on disease risk.
Novel insights into the molecular mechanisms of HLA class I abnormalities
  • B. Seliger
  • Biology
    Cancer Immunology, Immunotherapy
  • 2011
TLDR
A direct link between altered transcription factor binding, interferon signal transduction and MHC class I APM component expression has been shown, which will help to design effective T cell-based immunotherapies and give novel insights into the (patho) physiology of the antigen-processing and presenting pathway.
Upregulation of antigen-processing machinery components at mRNA level in acute lymphoblastic leukemia cells after CD40 stimulation
TLDR
Assessment of antigen-processing machinery (APM) components in acute lymphoblastic leukemia (ALL) cells before and after CD40 stimulation at messenger RNA (mRNA) level confirms specific stimulation of the antigen- processing system in ALL cells by CD40L.

Functional Non-equivalence of ATP-binding Cassette Signature Motifs in the Transporter Associated with Antigen Processing (TAP)*
TLDR
Investigation of the role of the ABC signature motif (C-loop) on the functional non-equivalence of the NBDs of TAP proves that the serine of the C-loop is not essential for TAP function but rather coordinates, together with other residues of the B-loop, the ATP hydrolysis in both nucleotide-binding sites.
Distinct Functions of the ATP Binding Cassettes of Transporters Associated with Antigen Processing
TLDR
A model in which nucleotide and peptide release from TAP are coupled and followed by ATP binding to TAP2, which induces high peptide affinity and initiates the transport cycle is proposed.
Identification of a contact region for peptide on the TAP1 chain of the transporter associated with antigen processing.
TLDR
It is concluded that the peptide binding site controls the selectivity of TAP and is composed of domains of both TAP1 and TAP2, which each contact the bound peptide over all or most of its length.
The ABC‐transporter signature motif is required for peptide translocation but not peptide binding by TAP
TLDR
It is concluded that the signature motifs in both TAP1 and TAP2 are required after peptide binding to facilitate peptide translocation by TAP.
Immune escape of melanoma: first evidence of structural alterations in two distinct components of the MHC class I antigen processing pathway.
TLDR
This study describes for the first time mutations in two distinct components of the MHC class I antigen processing pathway, suggesting an immune selection against CTLs recognizing both TAP-dependent and -independent T-cell epitopes.
A functionally defective allele of TAP1 results in loss of MHC class I antigen presentation in a human lung cancer
TLDR
This study evaluated 79 human solid tumours and cell lines for genetic abnormalities in TAP1 that might have led to an acquired loss of antigen presenting ability and found the first evidence for a naturally occuring protein structural defect resulting in defective peptide transport in a humansolid tumour.
Walker A Lysine Mutations of TAP1 and TAP2 Interfere with Peptide Translocation but Not Peptide Binding*
TLDR
An impairment in nucleotide hydrolysis by TAP complexes containing either mutant TAP subunit is suggested and the presence of one intact TAP NBD is insufficient for efficient catalysis of peptide translocation is indicated.
The “LSGGQ” Motif in Each Nucleotide-binding Domain of Human P-glycoprotein Is Adjacent to the Opposing Walker A Sequence*
TLDR
Results indicate that the signature sequences in P-gp likely participate in forming the ATP-binding sites and are displaced upon ATP hydrolysis, and may be the signal responsible for coupling ATP Hydrolysis to drug transport by inducing conformational changes in the transmembrane segments.
MHC class l/β2-microglobulin complexes associate with TAP transporters before peptide binding
TLDR
The results suggest that calnexin mediates class I/β2m dimerization, and subsequent binding of the dimers to TAP molecules facilitates their association with TAP-transported peptides.
