Oleocanthal, one of the minor components of the phenolic fraction isolated from extra virgin olive oil, is an effective inhibitor of COX enzymes, possessing similar potency to the NSAID ibuprofen. Moreover, it has the capacity to alter the structure of neurotoxic Aβ-amyloid oligomers (ADDLs) and to change the structure of deformed microtubule-associated tau–protein, thus inhibiting the formation of neurofibrillary tangles. It can have, therefore, potential therapeutic use for the treatment of…
Biomimetic Synthesis of Oleocanthal, Oleacein, and Their Analogues Starting from Oleuropein, A Major Compound of Olive Leaves.
TLDR
The method is stereocontrolled and provides an efficient approach to the synthesis of various oleocanthal analogues; thus, a small library of four compounds was prepared with 35-45% overall yield.
One-step semisynthesis of oleacein and the determination as a 5-lipoxygenase inhibitor.
TLDR
By exploiting oleuropein isolated from the plant leaf extract, an efficient procedure has been developed for a one-step semisynthesis of oleacein under Krapcho decarbomethoxylation conditions, facilitating the further development as a potential anti-inflammatory agent.
The Biological Activities of Oleocanthal from a Molecular Perspective
TLDR
Oleocanthal may act together with other bioactive compounds in olive oil to achieve its therapeutic potential and the use of oleOCanthal alone as a single therapeutic measure awaits validation from future studies.
Preparative isolation of oleocanthal, tyrosol, and hydroxytyrosol from olive oil by HPCCC.
TLDR
The equilibrium of (3S,4E)- and (3s,4Z)-OLC in such olive oil extracts has unambiguously been proven for the first time.
Health-promoting properties of oleocanthal and oleacein: Two secoiridoids from extra-virgin olive oil
TLDR
The aim of this review is to shed light on OLC and OLE, focusing on their precursors in the olive fruit and the impact of agronomic and processing factors on their presence in EVOO.
Semi-synthesis as a tool for broadening the health applications of bioactive olive secoiridoids: a critical review.
TLDR
The aim of this review is to summarize the semi-synthetic protocols published in the last fifteen years, covering 2005 up to 2020, which can produce natural olive bioactive phenols scarcely available by extractive procedures, and new biophenol derivatives with enhanced biological activity.
First total synthesis of (±)-floribundane B: An approach based on Johnson-Claisen rearrangement of a Morita-Baylis-Hillman adduct
Abstract The first total synthesis of (±)-floribundane B is reported. Johnson-Claisen rearrangement of a Morita-Baylis-Hillman adduct assembled all of the stereochemical features of this secoiridoid.
Olive oil oleocanthal and estrogen receptor expression
Abstract Oleocanthal is a naturally occurring phenolic compound in olive oil. Oleocanthal is the main component responsible for throat irritation and pungency of olive oil. The compound has
Elaboration of extra-virgin olive oils rich in oleocanthal and oleacein: pilot plant’s proposal
Extra-virgin olive oil is basically used in food, but it can also be used as a source of high added-value components, such as oleocanthal and oleacein. Oleocanthal and oleacein are bioactive
Anticancer molecular mechanisms of oleocanthal
TLDR
This review summarizes the current advancers concerning the anticancer activity of OLC, with particular emphasis on the molecular signaling pathways modulated by this compound in different tumor cell types.

Syntheses of (-)-oleocanthal, a natural NSAID found in extra virgin olive oil, the (-)-deacetoxy-oleuropein aglycone, and related analogues.
TLDR
The first- and now second-generation syntheses of both enantiomers of the oleocanthals, as well as the first synthesis of the closely related (-)-deacetoxy-oleuropein aglycone and a series of related analogues for structure activity studies are disclosed.
Phytochemistry: Ibuprofen-like activity in extra-virgin olive oil
TLDR
It is shown that newly pressed extra-virgin olive oil contains oleocanthal, a compound whose pungency induces a strong stinging sensation in the throat, not unlike that caused by solutions of the non-steroidal anti-inflammatory drug ibuprofen, which is an indicator of a shared pharmacological activity.
Synthesis and assignment of absolute configuration of (-)-oleocanthal: a potent, naturally occurring non-steroidal anti-inflammatory and anti-oxidant agent derived from extra virgin olive oils.
TLDR
Effective total syntheses and the assignment of absolute configurations of both the (+)- and (-)-enantiomers of oleocanthal 1 have been achieved and both enantiomers proved to be non-steroidal anti-inflammatory and anti-oxidant agents.
New insights on the interaction mechanism between tau protein and oleocanthal, an extra-virgin olive-oil bioactive component.
TLDR
A detailed mass spectrometric investigation of the OLC reactive profile with both tau protein fibrillogenic fragment K18 and propylamine in biomimetic conditions shows that K18 is prone to be covalently modified by OLC through Schiff base formation between the ε-amino group of lysine residues and OLC aldehyde carbonyls.

TLDR
Findings provide experimental support for the potential reduced risk of AD and related neurodegenerative diseases associated with olive oil consumption and may offer a new chemical scaffold for the development of AD-modulating agents.

Antioxidant and other biological activities of phenols from olives and olive oil
TLDR
Findings demonstrate that olive oil phenolics are powerful antioxidants, both in vitro and in vivo, and possess other potent biological activities that could partially account for the observed healthful effects of the Mediterranean diet.
Secoiridoids, tocopherols, and antioxidant activity of monovarietal extra virgin olive oils extracted from destoned fruits.
TLDR
It was concluded that a better knowledge of the reactions occurring during olive processing, and particularly on the involvement of endogenous pulp and stone enzymes, is essential to predict the effect of destoning on EVOO quality.
