In some depressive patients, regarded as therapy-resistant, the antidepressive effect of 1-5-hydroxytryptophan (in combination with a peripheral decarboxylase inhibitor) was ehanced by clomipramine at a dosage which, as such, was suboptimal. This probably means that the therapeutic effect of 1-5-hydroxy-tryptophan is based on increase of serotonin at central serotonin receptors.

Adjunctive 5-Hydroxytryptophan Slow-Release for Treatment-Resistant Depression: Clinical and Preclinical Rationale.
TLDR
Adjunctive administration of 5-hydroxytryptophan SR safely elevates 5-HTExt beyond the SERT inhibitor effect in humans; however,5-HTP cannot be a clinically viable drug because of its poor pharmacokinetics. Expand

In 1958 a new therapy for depressions was introduced: medication with antidepressants, with an effect on mood regulation which in suitable cases was primary and lasting, represented a new pharmacothera-peutic principle. Expand
The treatment of depression with L-5-Hydroxytryptophan versus imipramine
TLDR
The L-5-HTP results showed no difference compared with the results of an Imipramine treatment in 40 patients in earlier double-blind studies, and the gastrointestinal side effects seemed to be dose dependent. Expand
5-Hydroxytryptophan : A Clinically-Effective Serotonin Precursor by Timothy

CORRELATION BETWEEN CLINICAL EFFECTS OF VARIOUS ANTIDEPRESSANT TREATMENTS AND THEIR EFFECTS ON MONOAMINERGIC RECEPTORS IN THE BRAIN
TLDR
Antidepressant drugs which by various modes of action facilitate neurotransmission in central 5-hydroxytryptamine (5HT) neurons were investigated and their ability to activate 5HT receptors was evaluated by examining their effects on behaviour in mice. Expand
Safety of 5-hydroxy-L-tryptophan.
TLDR
Extensive analyses of several sources of 5-HTP have shown no toxic contaminants similar to those associated with L-Trp, nor the presence of any other significant impurities, which has raised undue speculations concerning its chemistry and toxicity. Expand

TLDR
The physiological role of 5-HTP in the treatment of depression, anxiety, panic, sleep disorders, obesity, myoclonus and serotonin syndrome are discussed and the occurrence of toxic impurities present in tryptophan and 5- HTP preparations are discussed. Expand
