Approximately 10% of all cases of breast cancer exhibit a familial pattern of incidence. Efforts to identify the genetic basis of familial breast cancer reached fruition some five years ago, when the breastcancer-susceptibility genes BRCA1 and BRCA2 were identified through positional cloning. Germline mutations in either of these genes account for 20-60% of breast cancer cases in families where multiple individuals are affected (~2-6% of all cases) (Nathanson et al., 2001).
