Triptolide, the main active ingredient in Tripterygium wilfordii Hook. f. (Celastraceae), has shown promising effects against a variety of tumors. However, the molecular pharmacological mechanisms explaining the action of triptolide remain unknown. In this study, the CT26 colon tumor cell line was inoculated subcutaneously into BALB/c mice, and plasma samples were subjected to 1H NMR metabolomics analysis. The metabolic signature identified five metabolites whose levels were lower and 15 whose…

Triptolide synergistically enhances antitumor activity of oxaliplatin in colon carcinoma in vitro and in vivo.
The result revealed that the combination therapy with TPL and OXA exerted synergistic antitumor effects at low concentration in colon cancer cells, with less cytotoxity, which exhibited high potency for clinical applications.
Triptolide induces protective autophagy through activation of the CaMKKβ-AMPK signaling pathway in prostate cancer cells
The results show that triptolide induces protective autophagy through the CaMKKβ-AMPK pathway in PCa cells, implying that a combination of tripsolide with autphagy inhibitors may potentially be an effective therapeutic strategy for PCa.
Triptolide inhibits cell proliferation and tumorigenicity of human neuroblastoma cells
Exposure of BE(2)-C human neuroblastoma cells to triptolide resulted in a reduction in cell growth and proliferation, and the induction of cell death and apoptosis, together with cell cycle arrest in the S phase.

The results indicate that MRx102 has potent antitumor effects both in vitro and in vivo, and is a potential novel therapy for the treatment of NSCLC.
Triptolide and its prodrug minnelide suppress Hsp70 and inhibit in vivo growth in a xenograft model of mesothelioma
Findings support the clinical evaluation of minnelide therapy for mesothelioma, which is a devastating disease with a poor prognosis for which there is a clear need for more successful therapeutic approaches.
