系统地研究了纯硅纳米颗粒(SiNP)、磷酸化硅纳米颗粒(PO4NP)和氨基化硅纳米颗粒(NH2NP)与人皮肤角质形成细胞系(HaCaT)的生物效应. 考查了3种不同功能化基团修饰的硅纳米颗粒对HaCaT细胞的细胞黏附效率、细胞增殖及细胞周期的影响, 以及HaCaT细胞对SiNP, PO 4 NP和NH 2 NP的吞噬情况. 结果表明: SiNP, PO 4 NP以及NH 2 NP 3种颗粒对HaCaT细胞的影响均存在浓度依赖关系, 当3种不同功能化基团修饰的硅纳米颗粒在细胞培养液中的终浓度低于0.2 μg/μL时, 与HaCaT细胞具有良好的生物相容性, 但是随着纳米颗粒在细胞培养液中终浓度的增大, PO 4 NP, SiNP和NH 2 NP对HaCaT细胞的影响也逐渐增大, 其中PO 4 NP所产生的影响随浓度增大的趋势最慢, SiNP次之, NH 2 NP最快; 同时, HaCaT细胞对纳米颗粒的吞噬量和吞噬速度也因其表面修饰的不同而存在差异, 在同样的作用浓度和作用时间下, NH 2 NP进入到细胞的量最多、速度最快, SiNP次之, PO 4 NP…
