Circulating IL-6 levels are elevated in obesity. Although IL-6 is expressed in adipose tissue, neither its regulation nor cell of origin is well characterized. Here we investigated the β-adrenergic regulation of IL-6 release in a combination of studies on humans and animals in vivo and cultured adipocytes in vitro. Human in vivo study: Human volunteers were infused with isoproterenol, norepinephrine, or saline {4 m:4F; mean (sd) age 35.5 (5.8) yr; body mass index 24.6 (4.2) kg/m−2}. Plasma IL-6…

Figures and Tables from this paper

Angiotensin II hypertension is attenuated in interleukin-6 knockout mice.

TLDR
The approximately 30 mmHg greater ANG II hypertension in the WT mice suggests that IL-6 contributes significantly to ANG II-salt hypertension, and early separation in MAP, the albumin excretion data, and the rapid, post-ANG II recovery of MAP suggest an IL- 6-dependent mechanism that is independent of renal injury.

Reduction of Plasma sCD40L and Stimulated MIP-1-Alpha Production by in vivo Beta-Adrenergic Stimulation

TLDR
The findings suggest that adipose tissue is a contributing source of β-adrenergically stimulated increases in circulating IL-6, and that the main source may not be PBMCs.

Cytokines in Metabolic Functions

TLDR
It is found that systemic LIF treatment could reduce white and brown fat depots in ovariectomized mice, suggesting that LIF can reduce obesity independently of estrogen signalling, and identified an association between an IL6ST/gp130 polymorphism in amino acid 148 (Gly/Arg) and risk of myocardial infarction in a hypertensive population.

TSH stimulates IL-6 secretion from adipocytes in culture.

TLDR
To the Editor: Adipocytes secrete interleukin-6 (IL-6), a pro-atherogenic cytokine, which is an independent cardiovascular disease (CVD) risk factor and hypothesized that TSH stimulates adipocyte IL-6 secretion.

Interleukin-6 induces impairment in human subcutaneous adipogenesis in obesity-associated insulin resistance

TLDR
Obesity-associated insulin resistance is marked by impaired SC adipogenesis, mediated, at least in a subset of individuals, by elevated local levels of IL-6, and understanding the molecular mechanisms underlying reduced adipogenic capacity in IR individuals could help target appropriate therapeutic strategies.

Role of endogenous IL-6 in the neonatal expansion and functionality of Wistar rat pancreatic alpha cells

TLDR
Data point to a potential role for local IL-6 in the regulation of alpha cell growth and function during suckling, and undernourished neonates showed altered alpha cell number and function and delayed appearance of IL- 6 in the pancreas.

The role of the brown adipose tissue in β3-adrenergic receptor activation-induced sleep, metabolic and feeding responses

TLDR
Findings indicate that the activation of BAT via β3-AR leads to increased sleep in mice and that this effect is dependent on the presence of UCP-1 protein and sleep responses require the intact sensory innervation of BAT.

Reduced stress- and cold-induced increase in energy expenditure in interleukin-6-deficient mice.

TLDR
The present results indicate that endogenous IL-6 is of importance for stress- and cold-induced energy expenditure in mice.

Endocrinologic and metabolic effects of interleukin-6 in humans.

TLDR
It is demonstrated that rhIL-6 induces many of the endocrinologic and metabolic changes found in catabolic states and thus may mediate some of the metabolic effects previously ascribed to other cytokines.

Subcutaneous adipose tissue releases interleukin-6, but not tumor necrosis factor-alpha, in vivo.

TLDR
Although both IL-6 and TNF alpha are expressed by adipose tissue, the results show that there are important differences in their systemic release.

Dose-dependent effects of recombinant human interleukin-6 on glucose regulation.

TLDR
Sc IL-6 administration induced dose-dependent increases in fasting blood glucose, probably by stimulating glucagon release and other counteregulatory hormones and/or by inducing peripheral resistance to insulin action.

The differential effect of food intake and beta-adrenergic stimulation on adipose-derived hormones and cytokines in man.

TLDR
Data show that secretion of some, but not all, metabolically relevant polypeptides by adipose tissue is modulated within a short time frame by food or stress stimuli, suggesting a role of these peptides in local autocrine/paracrine or distant endocrine effects on fat metabolism.

Induction of plasma interleukin-6 by circulating adrenaline in the rat

Dose effects of recombinant human interleukin-6 on pituitary hormone secretion and energy expenditure.

TLDR
Subcutaneous IL-6 administration induced synchronized dose-dependent increases in the RMR and hypothalamic-pituitary-adrenal axis activity, suggesting that hypothalamic corticotropin-releasing hormone may mediate both of these functions in humans.

Human breast adipocytes express interleukin-6 (IL-6) and its receptor system: increased IL-6 production by beta-adrenergic activation and effects of IL-6 on adipocyte function.

TLDR
The secretion of IL-6 by adipocytes is strongly stimulated by beta-adrenergic activation and is modestly suppressed by glucocorticoids, suggesting an autocrine/paracrine role of this cytokine in human adipose breast tissue.

Interleukin-6 secretion in mice is associated with reduced glucose-6-phosphatase and liver glycogen levels.

TLDR
Secretion of IL-6 from the developed tumors was associated with decreased food consumption, reduced body weight, and reduced blood glucose levels, and inhibition of G-6-Pase gene transcriptional rate was markedly reduced.

Interleukin-3 and interleukin-6 stimulate cortisol secretion from adult human adrenocortical cells.

TLDR
IL-3 and IL-6 directly stimulate the steroidogenesis at the adrenal level through activation of different, cAMP-independent pathways, and the effect of IL-3 on adrenocortical cortisol secretion is dependent on the lipoxygenase pathway.
