Abstract: Recent data from several groups suggest that the primary mechanism of β‐amyloid neurotoxicity may be mediated by reactive oxygen species. To evaluate this hypothesis, we first compared the efficacy of antioxidant agents in preventing toxicity caused by oxidative insults (iron, hydrogen peroxide, and tert‐butyl hydroperoxide) and β‐amyloid peptides in cultured rat hippocampal neurons. Tested antioxidants (propyl gallate, Trolox, probucol, and promethazine) generally provided…
Pyruvate protection against β‐amyloid‐induced neuronal death: Role of mitochondrial redox state
TLDR
It is found that P/M addition attenuated cell death evoked by β‐amyloid peptides after 24 hr treatment and that this effect was blocked by α‐ciano‐3‐hydroxycinnamate (CIN), suggesting that it requires mitochondrial pyruvate uptake.
Overexpression of Glutathione Peroxidase Increases the Resistance of Neuronal Cells to Aβ‐Mediated Neurotoxicity
TLDR
The hypothesis of a role of H2O2 in the mechanism of Aβ toxicity is strengthened and the potential of Ad‐GPx to reduce Aβ‐induced damage to neurons is highlighted, which may have applications in gene therapy for AD.
Amyloid β-peptide(1–40)-mediated oxidative stress in cultured hippocampal neurons
TLDR
It is reported that neurons, subjected to A β(1–40), respond to Aβ oxidative impact by activation of antioxidant defense mechanisms and alternative ATP-regenerating pathway and the maintenance of a constant level of CK function by increased CK BB production together with the induction of antioxidant enzyme gene expression in Aβ-treated hippocampal neurons.
Iron toxicity in organotypic cultures of hippocampal slices: role of reactive oxygen species
TLDR
It is indicated that iron elicits cellular damage predominantly by oxidative stress, and that ROS‐mediated iron toxicity may involve cytochrome c‐ and caspase‐3‐dependent apoptotic pathways.
Nicotine attenuates β‐amyloid peptide‐induced neurotoxicity, free radical and calcium accumulation in hippocampal neuronal cultures
TLDR
The present study demonstrates that nicotine protected cultured hippocampal neurons against the Aβ‐induced apoptosis, and suggests that nicotine may be beneficial in retarding the neurodegenerative diseases such as AD.
Caspase-2 mediates neuronal cell death induced by beta-amyloid.
TLDR
In all of the neuronal populations studied here, cell death was blocked by the broad spectrum caspase inhibitor N-benzyloxycarbonyl-val-ala-asp-fluoromethyl ketone and more specifically by the downregulation of casp enzyme-2 with antisense oligonucleotides, suggesting that formation of peroxynitrite is not critical for Abeta-mediated death.
Caspase-2 Mediates Neuronal Cell Death Induced by β-Amyloid
TLDR
The results indicate that caspase-2 is necessary for Aβ1–42-induced apoptosis in vitro, and neurons from casp enzyme-2 null mice were totally resistant to Aβ 1–42 toxicity, confirming the importance of this caspases in Aβ-induced death.
