Enzymes of the cyclooxygenase pathways of prostanoid biosynthesis.
This chapter reviews the biochemistry and the biochemical pharmacology of the enzymes involved in the formation of prostanoids and investigates the role of phospholipase A2 and PLA2 in regulating prostanoid biosynthesis. Expand

IRL 1620 (10(-9)-10(-7) M) induced contractions of the guinea pig trachea with a comparable potency to those of ET-1 or ET-3, suggesting that IRL 1520 is a potent ETB receptor agonist. Expand
Adenosine A2A, but not A1, receptors mediate the arousal effect of caffeine
It is reported that caffeine at 5, 10 and 15 mg kg−1 increased wakefulness in both wild-type mice and A1 receptor knockout mice, but not in A2A receptor knockout mouse, indicating that caffeine-induced wakefulness depends on adenosine A2a receptors. Expand
Biochemical, structural, genetic, physiological, and pathophysiological features of lipocalin-type prostaglandin D synthase.
X-ray crystallographic analyses revealed that PGDS possesses a beta-barrel structure with a hydrophobic pocket in which an active thiol, Cys(65), the active center for the catalytic reaction, was located facing to the inside of the pocket. Expand
The endogenous somnogen adenosine excites a subset of sleep-promoting neurons via A2A receptors in the ventrolateral preoptic nucleus
The present study is the first demonstration of a direct activation of the sleep neurons by adenosine, and further support the cellular and functional heterogeneity of theSleep neurons, which could enable their differential contribution to the regulation of sleep. Expand
Prostaglandin D2 as a mediator of allergic asthma.
PGD2 functions as a mast cell-derived mediator to trigger asthmatic responses and is investigated by the generation of mice deficient in the PGD receptor (DP). Expand
Prostaglandin D synthase: structure and function.
Two distinct types of PGD synthase are purified; one is the lipocalin-type enzyme and the other is the hematopoietic enzyme, which acts as a PGD2-producing enzyme and also as a lipophilic ligand-binding protein, with high affinities. Expand

It is shown that aβ-sheet conformational transition of the expanded polyQ protein monomer precedes its assembly into β-sheet–rich amyloid-like fibrils and is a therapeutic target for polyQ diseases and possibly for conformational diseases in general. Expand
The PGD2 pathway, independently of FGF9, amplifies SOX9 activity in Sertoli cells during male sexual differentiation
It is demonstrated genetically that the L-Pgds/PGD2 pathway acts as a second amplification loop of Sox9 expression, which suggests that, once activated and maintained by SOX9, production of testicular L-PGDS leads to the accumulation of PGD2, which in turn activates Sox9 transcription and nuclear translocation ofSOX9. It is established that the 26-kDa fertility-associated protein in bull seminal plasma is lipocalin-type PGD synthase, and it is speculated that this protein may play an important role in both the development and the maturation of sperm. Expand
