The dapE gene of Escherichia coli encodes N-succinyl-L-diaminopimelic acid desuccinylase, an enzyme that catalyzes the synthesis of LL-diaminopimelic acid, one of the last steps in the diaminopimelic acid-lysine pathway. The dapE gene region was previously purified from a lambda bacteriophage transducing the neighboring purC gene (J. Parker, J. Bacteriol.… (More)
Bell Laboratories digital computers were used during World War II principally in the design and testing of electrical analog gun directors. These computers evolved from the pioneer work of George R. Stibitz (in the period from 1937 to 1940), who designed and put to work at Bell Laboratories the first electromechanical digital computer.
The Escherichia coli dapF gene was isolated from a cosmid library as a result of screening for clones overproducing diaminopimelate epimerase. Insertional mutagenesis was performed on the cloned dapF gene with a mini-Mu transposon, leading to chloramphenicol resistance. One of these insertions was transferred onto the chromosome by a double-recombination… (More)
TGFbeta1 acts as a potent negative regulator of the cell cycle and tumor suppressor in part through induction of cyclin dependent kinase inhibitors p15(ink4b), p21, and p57. We previously showed that primary mouse epidermal keratinocytes (MEK) expressing a v-ras(Ha) oncogene undergo hyperproliferation followed by growth arrest and senescence that was… (More)
We present a novel type of ultrathin hydrogel microcapsules with pH-triggered shape switch. The capsules are produced as hollow hydrogel replicas of cubical inorganic templates and capable of keeping cubical geometries at neutral pH but transform into bulged structures at basic pH.
We have assessed the effects of five sulfonamides with widely varying inhibitory activity for carbonic anhydrase (CA) in the bone slice assay using disaggregated rat osteoclasts (OCs), and in the Maren assay where the catalytic activity of purified CA isozyme II (CA II) was measured. There was an excellent correlation between the relative potencies of the… (More)
The biological activity of various cigarette smokes has been assessed by treating cell cultures of rabbit alveolar macrophages with salt solutions of "whole smoke," smoke "vapor phase," and smoke "particulate phase." The results of these tests revealed degrees of toxicity that varied in accordance with the smoke treatment used. Further, these differences in… (More)
Recombinant diaminopimelate epimerase is overproduced to give 1% of soluble protein when grown under the appropriate conditions in Escherichia coli. This compares with 0.02% of the constitutive level of wild-type enzyme. A new purification procedure now yields milligram quantities of homogeneous enzyme of high specific activity (192 U/mg). This has enabled… (More)
