The nuclear RNase III Drosha initiates microRNA processing
- Yoontae Lee, C. Ahn, V. Kim
- BiologyNature

The two RNase III proteins, Drosha and Dicer, may collaborate in the stepwise processing of miRNAs, and have key roles in miRNA-mediated gene regulation in processes such as development and differentiation.
Regulation of microRNA biogenesis
Small non-coding RNAs that function as guide molecules in RNA silencing are involved in nearly all developmental and pathological processes in animals and their dysregulation is associated with many human diseases.
Biogenesis of small RNAs in animals
This Review summarizes the current knowledge of how these intriguing molecules are generated in animal cells.
MicroRNA genes are transcribed by RNA polymerase II
- Yoontae Lee, Minju Kim, V. Kim
- BiologyEMBO Journal
- 13 October 2004
The first direct evidence that miRNA genes are transcribed by RNA polymerase II (pol II) is presented and the detailed structure of a miRNA gene is described, for the first time, by determining the promoter and the terminator of mir‐23a∼27a‐24‐2.

These tiny, ∼22-nucleotide, RNAs control several pathways including developmental timing, haematopoiesis, organogenesis, apoptosis, cell proliferation and possibly even tumorigenesis.
Molecular Basis for the Recognition of Primary microRNAs by the Drosha-DGCR8 Complex

The Drosha-DGCR8 complex in primary microRNA processing.
- Jinju Han, Yoontae Lee, Kyu‐Hyun Yeom, Young‐Kook Kim, Hua Jin, V. Kim
- BiologyGenes & Development
- 15 December 2004
It is shown that DGCR8 may be an essential component of the pri-miRNA processing complex, along with Drosha, and a model for the action mechanism of class II RNase III proteins is proposed.

In vivo and in vitro evidence is presented that these clustered miRNAs are expressed polycistronically and are processed through at least two sequential steps, including the two processing steps and the nuclear export step, suggesting that the regulation of miRNA expression may occur at multiple levels.
The role of PACT in the RNA silencing pathway
- Yoontae Lee, Inha Hur, S. Park, Young‐Kook Kim, M. Suh, V. Kim
- BiologyEMBO Journal
- 8 February 2006
This study indicates that, unlike other RNase III type proteins, human Dicer may employ two different dsRBD‐containing proteins that facilitate RISC assembly, and describes an additional dsRNA‐binding protein known as PACT, which is significant in RNA silencing.
