The opportunistic marine pathogen Vibrio parahaemolyticus becomes virulent by acquiring a plasmid that expresses a deadly toxin
It is shown that an AHPND-causing strain of V. parahaemolyticus contains a 70-kbp plasmid with a postsegregational killing system, and that the ability to cause disease is abolished by the natural absence or experimental deletion of the plasmids-encoded homologs of the Photorhabdus insect-related toxins PirA and PirB. Mechanism of the Maturation Process of SARS-CoV 3CL Protease*[boxs]
It is reported here that the 3CLpro containing additional N- and/or C-terminal segments of the polyprotein sequences undergoes autoprocessing and yields the mature protease in vitro, and the dimeric three-dimensional structure of the C145A mutant protease shows that the active site of one protomer binds with the C- terminal six amino acids of the protomer from another asymmetric unit, suggesting a possible mechanism for maturation. The crystal structure of a left-handed Sulfolobus solfataricus RadA helical filament shows an evolutionarily conserved hinge region (a subunit rotation motif) in which a 360° clockwise axial rotation accompanies stepwise structural transitions from a closed ring to the AMP–PNP right- handed filament, then to an overwound right-handed filament and finally to the left-anded filament.
