Grape seed extract inhibits the growth and pathogenicity of Staphylococcus aureus by interfering with dihydrofolate reductase activity and folate-mediated one-carbon metabolism.
GSE might serve as an effective natural alternative for the control of food poisoning caused by S. aureus with proper safety measure, indicated that GSE-induced perturbation in folate-mediated one-carbon metabolism has more profound impact on methionine cycle than on thymidine monophosphate synthesis. Zebrafish larvae exposed to ginkgotoxin exhibit seizure-like behavior that is relieved by pyridoxal-5′-phosphate, GABA and anti-epileptic drugs
Ginkgotoxin-induced seizure in zebrafish larvae could serve as an in vivo model for epileptic seizure research and potential AED screening, and structural and morphological characterization showed that a 2-hour ginkgot toxin exposure did not cause appreciable changes in larval morphology and tissues development. Characterization and Comparative Studies of Zebrafish and Human Recombinant Dihydrofolate Reductases—Inhibition by Folic Acid and Polyphenols
The substantial resemblance between zebra fish and human DHFRs provides compelling evidence supporting the use of zebrafish DHFR as an in vitro assay system for folate-related studies and drug discovery. Expand

Folate deficiency-induced oxidative stress contributed to the folate deficiency-associated neuropathogenesis in both early and late stages of life, supporting the causal link between the increased oxidative stress and the folATE deficiency- induced abnormalities. Expand
Ethanol-Induced Upregulation of 10-Formyltetrahydrofolate Dehydrogenase Helps Relieve Ethanol-Induced Oxidative Stress
It is concluded that upregulation of fdh involving CEBPα helps relieve embryonic oxidative stress induced by ethanol exposure and helps relieve one-carbon homeostasis in ethanol-exposed embryos. Expand
Methotrexate-induced decrease in embryonic 5-methyl-tetrahydrofolate is irreversible with leucovorin supplementation.
It is found that tetrahydrofolate, the backbone of one-carbon carrier, gradually increased and became dominant in developing zebrafish embryos and methotrexate lowered the level of 5-CH3-THF in developing embryos, which could not be reversed with folate supplementation and might be more substantial to cellular methylation potential and epigenetic control than to nucleotide synthesis. Recombinant Zebrafish γ-Glutamyl Hydrolase Exhibits Properties and Catalytic Activities Comparable with Those of Mammalian Enzyme
This recombinant zγGH is active and exhibits comparable endopeptidase activity with folate substrate and antifolate drug methotrexate and significantly increased efficiency in folylpolyglutamate hydrolysis for folate analysis compared with current protocols. Expand
Knocking down 10-Formyltetrahydrofolate dehydrogenase increased oxidative stress and impeded zebrafish embryogenesis by obstructing morphogenetic movement.
Supporting the anti-oxidative activity of 10-formyltetrahydrofolate dehydrogenase and the in vivo function of tetrahyd rofolate conservation for 10- formylterahydofolate dehydrationase N-terminal domain is reported. Expand
Structures of the hydrolase domain of zebrafish 10-formyltetrahydrofolate dehydrogenase and its complexes reveal a complete set of key residues for hydrolysis and product inhibition
Structures of the hydrolase domain of 10-formyltetrahydrofolate dehydrogenase from zebrafish and its complexes are reported.
Structural insights into the hydrolysis and polymorphism of methotrexate polyglutamate by zebrafish γ-glutamyl hydrolase.
The first crystal structures of the endopeptidase-type γGH (zγGH) from zebrafish and the mutant complexes with MTX(Glu)5 and hydrolyzed MTX-1 are described, revealing the complete set of key residues involved in hydrolysis as well as the substrate-binding subsites. Expand
