Various sugars and their lactones were tested for their inhibition of beta-galactosidase (Escherichia coli). L-Ribose, which in the furanose form has a hydroxyl configuration similar to that of D-galactose at positions equivalent to the 3- and 4-positions of D-galactose, was a very strong inhibitor, and D-lyxose, which in the furanose form also resembles… (More)
The proton and carbon-13 NMR resonances for the 13-residue synthetic RRsrc peptide were completely assigned using two-dimensional NMR spectroscopy. This peptide contains a tyrosine in position 9 that can be phosphorylated by many tyrosine protein kinases. On the basis of observed nuclear Overhauser enhancements and alpha-proton and alpha-carbon chemical… (More)
A broad-specificity beta-glycosidase from porcine kidney was purified to homogeneity. Sodium dodecyl sulfate - polyacrylamide gel electrophoresis showed that it had a monomeric molecular weight of 55,000-60,000. Gel filtration showed native molecular weight of about 115,000. These data imply that the native enzyme is a dimer. The enzyme can catalyze the… (More)
The major form of the oligosaccharide of hen phosvitin was studied with two-dimensional 1H NMR of the intact glycoprotein. Its structure was determined from an analysis of the chemical shifts of the structural reporter groups, and it was further confirmed by comparison to several related model oligosaccharides. The oligosaccharide is N-linked and is present… (More)
