TMC-95A did not inhibit m-calpain, cathepsin L, and trypsin at 30 microM, suggesting its high selectivity for proteasome, and taxonomy of the producing strain is also described.

These results indicated that A. rugatobispora strains were distributed worldwide and were more resistant to 100 μg/ml gentamicin and 20 μg/ML ampicillin than Streptomyces strains, which interfere with the growth of A. Rugatob ispora selectively.

Four novel proteasome inhibitors, TMC-95A-D (1-4) have been isolated from the fermentation broth of Apiospora montagnei Sacc and the relationships of 1-4 to be diastereomers at C-7 and C-36 were revealed.
