Galetin 3,6-dimethyl ether (FGAL) is a flavonoid isolated from the aerial components of Piptadenia stipulacea (Benth) Ducke that shows a non-selective spasmolytic effect on each of the smooth muscle preparations the authors have tested, but with a greater effect on those from the rat aorta.

It is shown that, accordingly to the time of exercise, the rat ileum has its contractile reactivity reduced, in addition to the increased lipid peroxidation followed by its reestablishment to basal levels, as well as the augmented longitudinal smooth muscle layer thickness together with the reduced circular thickness.

The FGAL vasorelaxant mechanism involves noncompetitive antagonism of α1 adrenergic receptors, the non-selective opening of K+ channels, inhibition of Ca2+ influx through CaV or ROCs and the inhibition of intracellular Ca2+, particularly through PDE V inhibition.
Flavonoid galetin 3,6-dimethyl ether attenuates guinea pig ileum contraction through K(+) channel activation and decrease in cytosolic calcium concentration.

The main findings showed that the respiratory system works as a highly propitious place for the formation of ROS and RNS, especially superoxide anion, hydrogen peroxide, and peroxynitrite, and the epithelial damage is reflected in an important loss of antioxidant defenses that culminates in an imbalance and formation of inflammatory and contractile mediators.

It is concluded that the crude ethanol extract from S. paniculatum aerial parts presents antidiarrheal activity in mice related to the inhibition of small intestinal motility and secretion as well as nonselective spasmolytic activity on the guinea pig ileum.
