It is shown that SPOP serves as an adaptor of Daxx for the ubiquitination by Cul3-based ubiquitin ligase and subsequent degradation by the proteasome, and SPOP/Cul3-ubiquit in ligase plays an essential role in the control of Dxx level and, thus, in the regulation of DAXx-mediated cellular processes, including transcriptional regulation and apoptosis. It is shown that ISGylation played an essential role in the downregulation of ΔNp63α, and this findings establish ISG15 as a tumor suppressor via its conjugation to ΔN p63α and provide a molecular rationale for therapeutic use of doxorubicin against ΔNP63α-mediated cancers. Expand
