It is demonstrated that JTC‐801 antagonizes the ORL1 receptor response, and that J TC‐801 has efficacious and potent anti‐nociceptive effects in acute pain animal models not only by intravenous injection but also oral administration.

A search for sequences homologous to these ORFs found that the former product strongly resembles that of the 6-hydroxylation enzyme for oxytetracycline biosynthesis, and that the latter product has a weak but significant similarity to the hydroxyindole O-methyltransferase of bovine pineal gland.

The enzyme was strongly inhibited by Cu2+ ions, p-chloromercuribenzoate and N-bromosuccinimide, and also inhibited competitively by the reaction product, pyruvate, and its structurally related compounds, dihydroxyacetone and DL-glyceraldehyde.

In the analysis of the metal-substituted enzymes, it is found that the acid limb of pH rate profiles shifts from 7.2 for the original zinc enzyme to 6.6 for the cobalt enzyme, which suggests that the zinc atom is essential for the catalytic activity of the enzyme similarly to the zinc atoms in carboxypeptidase A.
