Production and characterization of monoclonal antibody to a melanoma specific glycoprotein.
Results demonstrate the utility of soluble antigen preparations devoid of strongly immunogenic non tumor-specific molecules in the elicitation of tumor specific antibody and suggest that immunogens of this kind are superior to intact melanoma cells for production of tumor Specific hybridomas.

Very large differences in the localization of radioactivity were observed in the thyroids and stomachs of mice when the iodobenzoyl-labeled antibodies were compared with the same antibodies labeled using the chloramine-T method of radioiodination.

A pattern of results suggested that the antiglobulin was a secondary immune reaction with elevation of the levels of preexisting antiglOBulin which was cross-reactive with the mouse antibody administered, and that this reaction was expected to present major complications to monoclonal antibody therapy.

In vivo experiments showed that gelonin conjugates are not toxic up to 2 mg total dose/mouse and significantly retarded the growth of established s.c. tumor.

It appears that a variety of parameters can affect ADCC reactions, including the type of effector cell and its level of activation, the isotype of the antibody, and properties of the target cell line such as its susceptibility to lysis.

The usefulness of immunofluorescent flow cytometry for quantitative assessment of antibody localization in solid tumors and provided information necessary for the design of further trials of monoclonal antibodies and immunoconjugates is demonstrated.

It is indicated that murine monoclonal antibodies have potential as selective targeting agents in the design of future therapeutic trials using monoclynal antibodies or conjugates thereof in the treatment of cancer.

Evaluation of cobalamin derivatives which were conjugated at the 5'-OH of ribose or the cobalt metal center indicated that conjugation at either of these positions had little effect on binding with rhTCII, however, conjugates where the attachment was made on the corrin ring substituents had a large variation in binding with RhTCII.
Kinetic model for the biodistribution of an 111In-labeled monoclonal antibody in humans.
The proposed nonlinear compartmental model predicts the dose levels required to overcome saturable background, suggests appropriate doses and schedules for cold loading strategies, and provides a format for explicit inclusion of tumor antigen.
