It is concluded that idazoxan may be used to distinguish alpha-1 and alpha-2 adrenoceptors on vascular smooth muscle in vitro and that the results favor the existence of alpha- 1 andalpha-2 adrenaline subtypes in terms of the existing subclassification scheme for alpha adrenoceptor subtypes.

The results in guinea-pig atria are consistent with a partial agonistic effect ofClonidine on prejunctional α-adrenoceptors; as an agonist clonidine is approximately 500 times more potent on these receptors than on postjunctionals α- adrenoceptor in rabbit aorta.

It is concluded that postjunctional alpha 2-adrenoceptors substantially mediate sympathetic vasoconstriction in distal segments of Sprague-Dawley rat tail artery and that this in vitro preparation may be a useful model for elucidating and extending data obtained in vivo in the human forearm circulation.
Influence of neuronal uptake on the contribution of smooth muscle α2-adrenoceptors to vasoconstrictor responses to noradrenaline in SHR and WKY isolated tail arteries
It appears that smooth muscle α2-adrenoceptors are present in SHR tail arteries, both intra- and extrajunctionally, and cocaine decreases the relative contribution of postjunctional α2 -adrenOceptors to responses to both exogenous and endogenous noradrenaline.

The results support the contention that postjunctional alpha 2-adrenergic receptors can be demonstrated in arterial smooth muscle in vitro and are particularly evident in arteries from hypertensive animals.

It is indicated that alpha-2 adrenoceptors predominate in both SHR and WKY tail arteries, but a significant subpopulation of smooth muscle alpha-1 adrenoceptor agonists is present in tail arteries of SHR but not of WKY rats.

The results obtained are consistent with the existence of presynaptic α‐ and β‐adrenoceptors on preganglionic terminals, and may be part of a trans‐synaptic inhibitory feedback mechanism; however the functional role of the facilitatory β‐ adrenoceptor is not clear.
