This is the first study concerning the enzymatic production of β-alanine by using ADC, and a pH–stat directed, fed-batch feeding strategy was developed to keep the pH value within 6–7.2 and thus attenuate substrate inhibition.

The RoAmy hydrolytic activity was the highest on wheat starch but showed only 55% activity on amylopectin relative to soluble corn starch, while the pullulanase activity was negligible, and the main end products of the polysaccharides tested were glucose and maltose.

Role of the calcium-binding residues Asp231, Asp233, and Asp438 of Bacillus amyloliquefaciens α-amylase (BAA) on the enzyme properties was investigated by site-directed mutagenesis and resulted in a dramatic reduction in the value of catalytic efficiency (Kcat/Km) and thermo-stability at 60°C.
