Transcription factor-based screens and synthetic selections for microbial small-molecule biosynthesis.
This work has developed a generalized approach to screen or select for improved small-molecule biosynthesis using transcription factor-based biosensors, and demonstrates product-dependent growth in E. coli using all three compounds.

It is demonstrated that mutant split T7 RNAP can be used as a transcriptional AND gate and a unique library of components for use in synthetic gene circuits is introduced.
Tuning the dynamic range of bacterial promoters regulated by ligand-inducible transcription factors
This work builds combinatorial sets of AraC-and LasR-regulated promoters containing −10 and −35 sites from synthetic and Escherichia coli promoters to build transcriptional logic gates regulated by two and three ligand-inducible transcription factors.
Predicting Transcriptional Output of Synthetic Multi-input Promoters.
The input-output relations of recently developed synthetic multi-input promoters are examined and two methods for predicting their behavior are described, which facilitate the design of synthetic circuits that utilize multi- input promoters.
Modular, Multi-Input Transcriptional Logic Gating with Orthogonal LacI/GalR Family Chimeras
This work demonstrates that complex multi-input transcriptional logic gating can be achieved through the use of ligand-inducible chimeric transcription factors assembled from the LacI/GalR family, and demonstrates that orthogonal DNA-binding domains and their cognate operators allow the coexpression of multiple, orthogona AND gates.
Synthetic biology platform technologies for antimicrobial applications.
This review highlights emerging synthetic biology platform technologies that are geared toward the generation of new antimicrobial therapies, diagnostics, and discovery channels and the development of paper-based, cell-free expression systems holds promise in promoting the clinical translation of molecular biology tools for diagnostic purposes.
Synthetic biology: the many facets of T7 RNA polymerase
Split T7 RNA polymerase provides new avenues for creating synthetic gene circuits that are decoupled from host regulatory processes—but how many times can this enzyme be split, yet retain function?
A synthetic system for asymmetric cell division in Escherichia coli
A synthetic genetic circuit for controlling asymmetric cell division in Escherichia coli in which a progenitor cell creates a differentiated daughter cell while retaining its original phenotype is described, which point the way toward the engineering of multicellular systems from prokaryotic hosts.
AlloRep: A Repository of Sequence, Structural and Mutagenesis Data for the LacI/GalR Transcription Regulators.
This work compared the noncovalent residue contact networks of the LacI/GalR subfamilies to design and experimentally validate an allosteric mutant of a synthetic LacI-GalR repressor for use in biotechnology.
Stable maintenance of multiple plasmids in E. coli using a single selective marker.
This work shows that large-scale plasmid-based synthetic gene circuits need not be limited by the use of multiple antibiotic resistance genes.
