The transitions in gene expression that occur during differentiation of in-vitro cultured bloodstream forms to procyclic forms are described, in which mRNAs with functions in particular pathways, or encoding components of protein complexes, show almost identical patterns of regulation.

Overexpression of TbUBP2 upregulated the levels of several mRNAs potentially involved in cell division, including the CFB1 mRNA, which encodes a protein with a cyclin F-box domain, and regulation was mediated by the 3′-untranslated region and involved stabilization of the mRNA.

It is demonstrated that the amount of FLAM3 and its localisation is dependent on ClpGM6 expression and vice versa, and that FAZ is a key regulator of trypanosome shape, with experimental perturbations being life cycle form dependent.

It is shown that specific depletion of PK50 or PK53 from bloodstream stage trypanosomes resulted in the rapid accumulation of cells with two nuclei and two kinetoplasts, indicating that cytokinesis was specifically inhibited and led to a deregulation of the cell cycle and cell death.

It is shown that early-eluting fractions represent different G1 subpopulations that progress through the cell cycle with distinct temporal profiles post-elutriation, as exemplified by the observation of the maturation of a second flagellar basal body in late G1 phase, DNA replication in S phase, and dimethylation of histone H3 in mitosis/cytokinesis.
