The SH2 and SH3 domain-containing protein GRB2 links receptor tyrosine kinases to ras signaling
Cargo of Kinesin Identified as Jip Scaffolding Proteins and Associated Signaling Molecules
Results demonstrate a direct interaction between conventional kinesin and a cargo, indicate that motor proteins are linked to their membranous cargo via scaffolding proteins, and support a role for motor proteins in spatial regulation of signal transduction pathways.
Guanine-nucleotide-releasing factor hSos1 binds to Grb2 and links receptor tyrosine kinases to Ras signalling
The results indicate that the Grbl/hSos1 complex couples activated EGF receptor to Ras signalling, and a synthetic 10-amino-acid peptide containing the sequence PPVPPR specifically blocks the interaction.
The Maguk protein, Pals1, functions as an adapter, linking mammalian homologues of Crumbs and Discs Lost
The data identifies a new multiprotein complex that appears to be evolutionarily conserved and likely plays an important role in protein targeting and cell polarity.
Direct interaction of two polarity complexes implicated in epithelial tight junction assembly
The identification of a biochemical and functional link between these two complexes that is mediated by Par6 and PALS1 is highlighted, highlighting a previously unrecognized link between protein complexes that are essential for epithelial polarity and formation of tight junctions.

The tight junction is an intracellular junctional structure that mediates adhesion between epithelial cells and is required for epithelial cell function and the establishment and maintenance of apico-basal polarity.

Using yeast 2-hybrid and protein coprecipitation approaches in vitro, it is shown that the neuronal adaptor proteins FE65 and mammalian Disabled bind to the cytoplasmic tails of LRP, LDL receptor, and APP, where they can potentially serve as molecular scaffolds for the assembly of cytosolic multiprotein complexes.
Phosphatidylinositol 3′‐kinase is activated by association with IRS‐1 during insulin stimulation.
It is concluded that the binding of tyrosyl phosphorylated IRS‐1 to the SH2 domains of p85 is the critical step that activates PtdIns 3′‐kinase during insulin stimulation.
Mammalian Crumbs3 is a small transmembrane protein linked to protein associated with Lin-7 (Pals1).
ERBIN: a basolateral PDZ protein that interacts with the mammalian ERBB2/HER2 receptor
It is suggested that ERBIN acts in the localization and signalling of ERBB2/HER2 in epithelia and belongs to a new PDZ protein family.
